Interaction of a Homologous Series of Amphiphiles with P-glycoprotein in a Membrane Environment-Contributions of Polar and Non-Polar Interactions

Summary: The transport of drugs by efflux transporters plays a crucial role in drug resistance development by cancer cells and pathogens. In this study, the interaction between a series of fluorescent amphiphiles and the efflux transporter P-glycoprotein (P-gp) was explored. The results showed that the binding parameters obtained were apparent and a new formalism was proposed to better understand the binding affinity of amphiphiles with P-gp. It was found that the polar interactions are essential for this interaction, and an increase in lipophilicity and amphiphilicity enhanced the association with the lipid bilayer, inhibiting the function of P-gp.

MemCross: Accelerated Weight Histogram method to assess membrane permeability

Summary: Passive permeation events across biological membranes play a crucial role in the pharmacokinetics of drugs. The accurate and rapid prediction of membrane permeation coefficients is important for drug discovery. The latest method, MemCross, based on the Accelerated Weight Histogram (AWH) protocol, is a cost-effective approach to estimate the potential of mean force (PMF) and diffusivity profiles of drug-like xenobiotics, providing a detailed description of molecule orientations and interactions. This method has been successfully applied to 12 xenobiotics and revealed a coherent structure-activity relationship.

Ligand's Partition to the Lipid Bilayer Should Be Accounted for When Estimating Their Affinity to Proteins

Summary: Ligand-protein interactions in complex media with lipids can influence the apparent affinity and should be considered when studying protein interactions. This study presents a framework to quantitatively characterize these interactions and explores the validity of commonly used approximations. The results show that ligand lipophilicity and lipid bilayer volume impact the apparent ligand-protein affinity, with exceptions for very polar ligands and ligands with supralinear increase in affinity with lipophilicity. The study also discusses important parameters for exploring the specificity of membrane proteins.

Modeling the Effect of Hydrophobicity on the Passive Permeation of Solutes across a Bacterial Model Membrane

Summary: Passive diffusion across biomembranes is an important permeation mechanism for drugs, including antibiotics, but it is often overlooked in drug uptake studies and requires further investigation. In this study, molecular dynamics simulations were used to investigate the influence of changes in molecular hydrophobicity on the permeability of a series of inhibitors of Pseudomonas aeruginosa's quorum sensing. The findings show that permeation across a membrane model is not correlated with the molecule's hydrophobicity. A simple model based on the difference in free energy profile outperforms the inhomogeneous solubility-diffusion model in predicting permeability, especially for hydrophobic molecules. The calculated differences in permeability do not explain differences in bacterio activity, but substantial differences in molecular orientation along the permeation pathway correlate with bacterio activity.
