Metformin and Insulin Resistance: A Review of the Underlying Mechanisms behind Changes in GLUT4-Mediated Glucose Transport

Rok Herman et al.

Summary: This review discusses the mechanisms of action of metformin, focusing on changes in insulin signaling pathway mediators, AMPK, GLUT4 translocation mediators, and the impact of posttranscriptional modifications. It aims to provide a comprehensive understanding of the interactions between metformin action, GLUT4 expression, GLUT4 translocation, and peripheral insulin sensitivity.

The Role of Glp-1 Receptor Agonists in Insulin Resistance with Concomitant Obesity Treatment in Polycystic Ovary Syndrome

Krzysztof Bednarz et al.

Summary: Insulin resistance plays a crucial role in the pathogenesis of polycystic ovary syndrome (PCOS) and interacts with hormonal abnormalities in a vicious circle. Defects in insulin receptor-related signaling pathways are enhanced by obesity and its features. Lifestyle intervention is important for managing excess weight in PCOS, while anti-obesity pharmacological agents are increasingly recognized for their role in achieving and maintaining weight loss.

Trends in insulin resistance: insights into mechanisms and therapeutic strategy

Mengwei Li et al.

Summary: The centenary of insulin discovery provides an important opportunity to transform diabetes from a fatal diagnosis to a manageable chronic condition. Insulin resistance, associated with obesity, diabetes, and other abnormalities, is caused by abnormal biological response to insulin stimulation. Recent studies suggest that dysregulated metabolic mediators released by adipose tissue contribute to insulin resistance. Although there is no specific medication for insulin resistance, lifestyle changes and pharmacological interventions have been used to improve insulin sensitivity.

Hepatic and cardiovascular safety of acarbose among type 2 diabetes patients with end-stage renal disease: A nationwide population-based longitudinal study

Wei-Hung Lin et al.

Summary: This study assessed the relationship between acarbose use and hepatotoxicity, cardiovascular disease, and mortality in patients with type 2 diabetes and end-stage renal disease. It found that acarbose use was associated with significantly lower risks of hepatic injury, composite cardiovascular events, and all-cause mortality, especially in patients with more severe diabetes or longer diabetes duration.

Cardiovascular Events with Finerenone in Kidney Disease and Type 2 Diabetes

Bertram Pitt et al.

Summary: Finerenone therapy improved cardiovascular outcomes in patients with type 2 diabetes and stage 2 to 4 CKD with moderately elevated albuminuria or stage 1 or 2 CKD with severely elevated albuminuria, compared to placebo. The risk of the primary outcome event was significantly lower in the finerenone group, primarily driven by a reduction in hospitalization for heart failure.

Molecular Determination of mirRNA-126 rs4636297, Phosphoinositide-3-Kinase Regulatory Subunit 1-Gene Variability rs7713645, rs706713 (Tyr73Tyr), rs3730089 (Met326Ile) and Their Association with Susceptibility to T2D

Rashid Mir et al.

Summary: The study found an association between the Mir126 rs4636297 and PIK3R1 SNPs (rs7713645 and rs3730089) with Type 2 diabetes. The minor allele of rs4636297 was associated with increased risk of T2D, while rs7713645 and rs3730089 also showed significant differences in genotype distribution between cases and controls. Further studies are needed to confirm these results with larger sample sizes and different populations, as well as to investigate the impact of these SNPs on PI3K regulatory subunit and catalytic activity.
