A weight of evidence approach for bioaccumulation assessment

To address uncertainty, a weight of evidence (WoE) approach is recommended. The Bioaccumulation Assessment Tool (BAT) provides a user-friendly platform for assessing bioaccumulation, including data evaluation templates (DETs) to evaluate the reliability of the data, consistent with international principles and promoting chemical policy objectives. It is an iterative and tiered approach that reduces unnecessary animal testing.

Targeted Per- and Polyfluoroalkyl substances (PFAS) assessments for high throughput screening: Analytical and testing considerations to inform a PFAS stock quality evaluation framework

Summary: To accelerate the evaluation of PFAS, a high-throughput framework was developed to assess the quality of screening stocks for 205 selected PFAS. A quality scoring system was created using mass spectrometry coupled with chromatography, providing a pass or fail notation based on observations during mass spectral examination. Results showed that 148 PFAS passed the quality assessment and could proceed to in vitro testing, while 57 PFAS scored failing primarily due to detection issues or confounding effects of the diluent (DMSO).

Plasma Protein Binding Evaluations of Per- and Polyfluoroalkyl Substances for Category-Based Toxicokinetic Assessment

Summary: New approaches using in vitro screening and in silico approaches, rely on in vitro toxicokinetic (TK) data to evaluate chemical substances. The evaluation of 71 PFAS compounds was conducted to understand their plasma protein binding (PPB) by ultracentrifugation with liquid chromatography-mass spectrometry analysis. Results showed that over half of the PFAS compounds had PPB exceeding 99.5%, and binding was lower for longer carbon chain length PFCAs. Bayesian modeling was used to provide uncertainty bounds for incorporation into TK modeling. This evaluation greatly expands our current knowledge and will aid in PFAS NAM development.

More contemporary approaches such as read-across, molecular dynamics, and protein modeling are proposed to accelerate the development of toxicity information on PFAS, as well as the need for precaution to protect human health given the known effects of certain PFAS compounds.
