Prevalence Estimates of Predicted Pathogenic COL4A3-COL4A5 Variants in a Population Sequencing Database and Their Implications for Alport Syndrome

Summary: This study estimated the frequencies of predicted pathogenic COL4A3-COL4A5 variants in populations without known kidney disease. The COL4A5 variants were found in at least one in 2320 individuals, and the predicted pathogenic heterozygous COL4A3 and COL4A4 variants affected one in 106 of the population. The results suggest the population frequencies for Alport syndrome need to be adjusted for individual variant penetrance and other factors.

Summary: The study conducted in northeastern Italy over 5.5 years showed that 22 males were confirmed to have a GLA gene variant, with an incidence of 1:7879 newborns. All positive patients were asymptomatic at the last visit, with plasma lyso-Gb(3) levels used for non-invasive monitoring.
