Safety and efficacy of ranolazine in hypertrophic cardiomyopathy: Real-world experience in a National Referral Center

Alessia Argiro et al.

Summary: This study assessed the efficacy and safety of ranolazine in real-world patients with hypertrophic cardiomyopathy (HCM). The results showed that ranolazine had a certain therapeutic effect in relieving angina and reducing the burden of ventricular arrhythmias. However, no symptomatic improvement could be observed in patients with heart failure associated with severe diastolic dysfunction.

High-Sensitivity Cardiac Troponin T Elevation in Hypertrophic Cardiomyopathy Is Associated With Ventricular Arrhythmias

Daniel R. Burczak et al.

Summary: This study investigated the relationship between high-sensitivity cardiac troponin T (hs-cTnT) levels and sudden cardiac death (SCD) in patients with hypertrophic cardiomyopathy (HCM). The results showed that hs-cTnT levels were associated with known risk factors for SCD and previous ventricular arrhythmias and appropriate implantable cardioverter-defibrillator shocks. However, this association was no longer present when sex-specific hs-cTnT cutoffs were used. Therefore, further research should use different hs-cTnT reference values by sex to determine whether an elevated hs-cTnT value is an independent risk factor for SCD in patients with HCM.

Rare and Common Genetic Variation Underlying the Risk of Hypertrophic Cardiomyopathy in a National Biobank

Kiran J. Biddinger et al.

Summary: This study assessed the contributions of rare and common genetic variations to the risk of hypertrophic cardiomyopathy (HCM) and found that both types of variations significantly contribute to HCM susceptibility and improve HCM risk prediction.

Common genetic variants and modifiable risk factors underpin hypertrophic cardiomyopathy susceptibility and expressivity

Andrew R. Harper et al.

Summary: The study identified 12 genome-wide significant susceptibility loci for HCM, showing a strong polygenic influence, especially for sarcomere-negative HCM. A genetic risk score influenced the odds of HCM and phenotypic severity in carriers of sarcomere variants. Mendelian randomization identified diastolic blood pressure as a key modifiable risk factor for sarcomere-negative HCM.

Shared genetic pathways contribute to risk of hypertrophic and dilated cardiomyopathies with opposite directions of effect

Rafik Tadros et al.

Summary: The study found a strong genetic correlation between hypertrophic and dilated cardiomyopathies and left ventricular traits, with opposing genetic effects in the two cardiomyopathies. The research also supported a causal association between increased left ventricular contractility and hypertrophic cardiomyopathy risk.

Molecular Genetic Basis of Hypertrophic Cardiomyopathy

A. J. Marian

Summary: Hypertrophic cardiomyopathy (HCM) is a genetic disease characterized by left ventricular hypertrophy and may lead to heart failure or sudden cardiac death. Pathogenic variants in genes encoding myocardial protein constituents, especially MYH7 and MYBPC3 genes, are the main causes of HCM. Advances in genetic testing and drug development have improved the therapeutic prospects for HCM.
