Ghost messages: cell death signals spread

Mingming Zhang et al.

Summary: Cell death is accompanied by the release of active or passive molecules that can signal to and regulate physio- or pathological events. This review summarizes the types of messages cells can spread while dying, their effects on target tissue/cells, and how these communications regulate physiological or pathological processes. The aim is to identify unresolved questions, formulate new hypotheses, and provide references for the search of novel diagnostic/therapeutic agents.

Osteoporosis: Mechanism, Molecular Target and Current Status on Drug Development

Hanxuan Li et al.

Summary: Osteoporosis is a pathological condition characterized by a loss of bone mass due to an imbalance in bone remodeling. Current treatments include anti-resorptive agents and anabolic drugs, but severe side effects and loss of efficacy may limit long term usage. Sequential and combinational use of current drugs, as well as development of novel drugs against new targets, offer alternative approaches for osteoporosis treatment.

Osteoclasts recycle via osteomorphs during RANKL-stimulated bone resorption

Michelle M. McDonald et al.

Summary: Research has shown that RANKL-stimulated osteoclasts can undergo fission to form osteomorphs, which are involved in the regulation of bone resorption and could be targeted for the treatment of skeletal diseases. Single-cell RNA sequencing revealed that osteomorphs are transcriptionally distinct from osteoclasts and macrophages, expressing non-canonical osteoclast genes associated with bone phenotypes when deleted in mice.

The global prevalence of osteoporosis in the world: a comprehensive systematic review and meta-analysis

Nader Salari et al.

Summary: This study conducted a systematic review and meta-analysis to investigate the global prevalence of osteoporosis. The results showed an overall prevalence of 18.3%, with higher rates in women (23.1%) compared to men (11.7%). Africa had the highest prevalence of osteoporosis at 39.5%.

Bone Mineral Density Loss and Fracture Risk After Discontinuation of Anti-osteoporotic Drug Treatment: A Narrative Review

Laura P. B. Elbers et al.

Summary: Long-term anti-osteoporotic drug therapy requires continuous monitoring and treatment, as discontinuation of therapy may lead to rapid increases in bone turnover markers and significant bone density loss, especially in patients treated with denosumab and romosozumab.

The Development of Molecular Biology of Osteoporosis

Yongguang Gao et al.

Summary: Osteoporosis is a major bone disorder affecting both men and women, primarily due to an imbalance in bone remodeling. Understanding the effects of molecules produced by different cells on bone can help identify new therapeutic targets and approaches for preventing and treating bone disorders.

Osteoclast-derived apoptotic bodies couple bone resorption and formation in bone remodeling

Qinyu Ma et al.

Summary: The study identified the molecular profile of apoptotic bodies (ABs) derived from osteoclasts at different differentiation stages, showing marked differences in protein cargo and high similarity to parental cells. Functionally, pOC-ABs promoted endothelial progenitor cell differentiation and endothelial cell formation, while mOC-ABs induced osteogenic differentiation of mesenchymal stem cells and facilitated osteogenesis via RANKL reverse signaling, highlighting the important biological roles of ABs in coupling bone formation with resorption during bone remodeling.
