Protein-Mediated Carotenoid Delivery Suppresses the Photoinducible Oxidation of Lipofuscin in Retinal Pigment Epithelial Cells

Summary: Protein-mediated delivery of zeaxanthin using BmCBP-ZEA can suppress photoinducible oxidative stress in RPE cells caused by irradiation of lipofuscin with intense white light. In this study, we used fluorescence lifetime imaging to observe the changes in lipofuscin granules in ARPE-19 cell culture before and after white light irradiation, with and without the addition of BmCBP-ZEA. We found that after irradiation, the average fluorescence lifetime of lipofuscin significantly increased, while the presence of BmCBP-ZEA at 200 nM concentration suppressed the increase in lipofuscin fluorescence lifetime, indicating approximately 35% inhibition of oxidative stress. This phenomenon indirectly demonstrates the effectiveness of protein-mediated carotenoid delivery into pigment epithelium cells.

Summary: Water-soluble carotenoproteins, found in many organisms, have the potential to be used as antioxidant nanocarriers in biomedical applications. However, the characterized carotenoproteins are limited in variety. This study focuses on a specific carotenoid-binding protein from Bombyx mori and analyzes its binding repertoire and potential as a carotenoid delivery module. The findings show that the protein can form complexes with various carotenoids and efficiently shuttle them to biological membrane models and other carotenoproteins.

Structural basis for the carotenoid binding and transport function of a START domain

Summary: In this study, the crystal structures of a carotenoid-binding protein (BmCBP) from silkworm Bombyx mori were determined, revealing the interaction between BmCBP and zeaxanthin (ZEA). The research showed structural differences in the carotenoid-binding cavity between BmCBP and human STARD3, explaining their distinct carotenoid-binding capacities. These findings provide important insights into the function of xanthophyll-binding proteins in the eye.

Summary: This study focuses on the photoprotection mechanism in cyanobacteria mediated by Orange Carotenoid Protein (OCP) and its C-terminal domain homologs (CTDHs), which play a crucial role in multidirectional carotenoid transfer. NMR techniques were used to analyze the structure of CTDH, revealing a largely equivalent structure in solution and in the crystal. These findings shed light on the dynamics of CTDH and provide a platform for further analysis of the holo-CTDH structure and the development of antioxidant nanocarriers for biomedical applications.
