Phenols in Pharmaceuticals: Analysis of a Recurring Motif

Kevin A. Scott et al.

Summary: Phenols and phenolic ethers are important molecular structures in nature and approved drugs. This study provides the first comprehensive compilation and analysis of the structures of U.S. FDA-approved molecules containing phenol or phenolic ether fragments. The research reveals the differences in physicochemical properties between phenols and phenol ethers, and their significance in drug development. Despite recent trends in increasing sp(3) centers in drug leads, phenols and phenolic ethers still have a significant presence in small-molecule drugs approved in 2020, indicating their special role in drugs and natural products.

Pyridine-Conjugated Pillar[5]arene: From Molecular Crystals of Blue Luminescence to Red-Emissive Coordination Nanocrystals

Xin-Yue Lou et al.

Summary: By employing a facile supramolecular self-assembling strategy, a luminescent molecular crystal and a Cu(I)-coordinated luminescent nanocrystal were simultaneously prepared using a conjugated pillar[5]arene macrocycle. The molecular crystal exhibited enhanced luminescence due to its conjugated structure and staggered packing mode, while the coordination nanocrystal showed well-defined crystalline structures and long-lifetime triplet state emission with distinct solvochromic features.

Pyridine Scaffolds, Phenols and Derivatives of Azo Moiety: Current Therapeutic Perspectives

Tehreem Tahir et al.

Summary: Synthetic heterocyclic compounds show great potential in fighting various diseases, with pyridines, phenolic compounds, and azo derivatives displaying excellent antimicrobial, antiviral, and other activities. The presence of nitrogen in the phenyl ring, hydroxyl substituents, and incorporation of a diazo group are crucial for the compounds' efficacy. Recent studies compared these compounds with existing drugs and new synthetic derivatives to explore therapeutic possibilities.

Palladium-catalyzed one-pot phosphorylation of phenols mediated by sulfuryl fluoride†

Yiyuan Zhang et al.

Summary: This study presents a general palladium-catalyzed procedure for the synthesis of phosphonates, phosphinates, and phosphine oxides from phenols mediated by sulfuryl fluoride. The method shows mild conditions, broad substrate scope, high functionality tolerance, and water insensitivity. The utility of this procedure is demonstrated through various applications including gram-scale synthesis, late-stage decoration of drugs and natural products, and on-DNA synthesis for a DNA-encoded library.
