Genomic determinants impacting the clinical outcome of mogamulizumab treatment for adult T-cell leukemia/lymphoma

Summary: By conducting molecular analysis on adult T-cell leukemia/lymphoma (ATL) patients, it was found that CCR4 alterations were associated with better clinical response and overall survival, while TP53, CD28, and CD274 alterations were associated with worse overall survival. This study contributes to the establishment of precision medicine using mogamulizumab in ATL patients.

A targeting delivery system for effective genome editing in leukemia cells to reverse malignancy

Summary: A dual-targeting CRISPR-Cas9 genome editing nanosystem was developed to reverse the malignancy of acute myeloid leukemia (AML) cells. The nanosystem effectively knocked out CXCR4 gene, leading to inhibited cell growth and migration. Moreover, this study provides an effective strategy to evaluate therapeutic efficiency through peripheral blood-based ex vivo studies.

Clinical Guidelines and New Molecular Targets for Cutaneous Lymphomas

Summary: Primary cutaneous lymphomas are heterogeneous lymphoproliferative disorders, with a need for more effective treatment options for advanced disease. The frequency of each subtype varies among different ethnic groups and the differences in clinical guidelines worldwide are important to consider. Current chemotherapy is temporarily effective, but strategies targeting tumor-specific molecules are being developed for improved treatment outcomes.

Different chemokines can preferentially activate different signaling pathways via the same receptor, a phenomenon known as biased agonism. The study shows that different chemokine receptors exhibit varying signaling outcomes, suggesting that their intrinsic abilities to couple with downstream signaling pathways play a role in differential agonism.

Lactate-induced M2 polarization of tumor-associated macrophages promotes the invasion of pituitary adenoma by secreting CCL17

Summary: The study found a positive association between lactate and PA invasion. Invasive PAs were found to have high infiltration of M2-like TAMs. Lactate secreted by PA cells promoted M2 polarization via mTORC2 and ERK signaling pathways, while activated TAMs secreted CCL17 to promote PA invasion through the CCL17/CCR4/mTORC1 axis. High CCL17 expression was associated with larger tumor size, greater invasion, and higher susceptibility to postoperative recurrence in human PAs.
