Cyclic and Linear Peptides Containing Alternate WW and RR Residues as Molecular Cargo Delivery Tools

Among them, the peptide [WWRR](5) showed the highest cellular uptake of cargo molecules, indicating its potential as an effective molecular transporter. Highly Retina-Permeating and Long-Acting Resveratrol/Metformin Nanotherapeutics for Enhanced Treatment of Macular Degeneration

Summary: We developed a nanomedicine strategy to create nanotherapeutics with high retinal permeability and sustained bioactive delivery. The nanotherapeutics were designed through aminolysis of resveratrol-encapsulated polycaprolactone nanoparticles, followed by the formation of amide linkages with a transacting activator of transcription cell penetrating peptide and metformin. In vitro and in vivo studies demonstrated the effectiveness of the nanotherapeutics in improving retinal permeability and targeting retinal diseases.

[(WR)8WKβA]-Doxorubicin Conjugate: A Delivery System to Overcome Multi-Drug Resistance against Doxorubicin

Summary: In this study, a cyclic peptide [(WR)(8)WK beta A]-Dox conjugate was successfully synthesized by conjugating Doxorubicin (Dox) with a cyclic peptide containing alternate tryptophan and arginine residues. Antiproliferative assays demonstrated that the conjugate exhibited higher activity than Dox alone, and it also showed strong inhibitory effects on Dox-resistant cells. Confocal microscopy images confirmed the ability of the conjugate and physical mixture of the peptide and drug to deliver Dox through an endocytosis-independent pathway. The stability of the conjugate was observed to be relatively short in human serum.
