Phage Therapy in Germany-Update 2023

Christian Willy et al.

Summary: Bacteriophage therapy shows promise in addressing global and German antibiotic resistance crises. The current situation in Germany regarding phage therapy and research is outlined. Critical shortcomings include a lack of clinical trials, regulation, and phages for clinical use. Phage therapy is being used on a limited number of patients as individual treatment trials.

Methicillin-Susceptible Staphylococcus aureus Biofilm Formation on Vascular Grafts: an In Vitro Study

Cristina Tello-Diaz et al.

Summary: The aim of this study was to quantify in vitro biofilm formation by methicillin-susceptible Staphylococcus aureus (MSSA) on the surfaces of different types of commonly used vascular grafts. The Dacron silver-triclosan and Omniflow II vascular grafts showed the greatest resistance to in vitro methicillin-susceptible Staphylococcus aureus biofilm formation compared to other vascular grafts.

Phage-Antibiotic Synergy Inhibited by Temperate and Chronic Virus Competition

Kylie J. Landa et al.

Summary: This study investigates the synergistic and competitive effects between antibiotics and viruses (temperate and chronic) on antibiotic treatment for bacterial infections. It finds that the combination of antibiotics and temperate viruses has a synergistic effect, but competition between temperate and chronic viruses inhibits the efficacy of antibiotics.

The antagonistic interactions between a polyvalent phage SaP7 and 13-lactam antibiotics on combined therapies

Dongxin Ma et al.

Summary: This study identified a polyvalent phage SaP7 that exhibited antagonistic effects on multiple drug-resistant bacteria. It is crucial to fully and cautiously evaluate the interactions between phages and antibiotics to avoid antagonistic impacts and failure of combination therapy.

Aminoglycoside Antibiotics Inhibit Phage Infection by Blocking an Early Step of the Infection Cycle

Larissa Kever et al.

Summary: Predation by phages is a major driver of bacterial evolution. This study reveals that aminoglycosides, a type of antibiotics produced by Streptomyces, have potent antiviral properties and can block an early step of phage infection. The supernatants from natural aminoglycoside producers also show broad inhibitory effects on phage propagation. Acetylation of the aminoglycoside antibiotic apramycin abolishes its antibacterial effect but retains its antiviral properties. These findings expand our understanding of aminoglycoside functions and suggest that they may provide protection against phage predation at the community level.

Eradication of Biofilm-Mediated Methicillin-Resistant Staphylococcus aureus Infections In Vitro: Bacteriophage-Antibiotic Combination

Razieh Kebriaei et al.

Summary: Bacterial biofilms are difficult to eradicate and can complicate infections. Phage+antibiotic combinations (PAC) may be more effective on biofilms than single agents. Our study demonstrated the effectiveness of phage Sb-1 on MRSA biofilms and suggested PAC regimens as adjuncts to antibiotics.

Evaluation of Bacteriophage-Antibiotic Combination Therapy for Biofilm-Embedded MDR Enterococcus faecium

Katherine Lev et al.

Summary: This study examined the synergy between lytic bacteriophages and antibiotics in treating multidrug-resistant Enterococcus faecium. The results showed that combinations of bacteriophages with daptomycin and other antibiotics had synergistic and bactericidal effects, while also preventing the emergence of resistance.

Antibiotic Exposure Leads to Reduced Phage Susceptibility in Vancomycin Intermediate Staphylococcus aureus (VISA)

Shawna McCallin et al.

Summary: In Vancomycin-intermediate Staphylococcus aureus (VISA), exposure to vancomycin may lead to cross-resistance to phage. VISA strains with intermediate levels of vancomycin resistance showed decreased sensitivity to phage. Serial passaging with vancomycin induced reduced vancomycin susceptibility and phage sensitivity. Understanding the interference between antibiotic and phage resistance mechanisms is important for designing effective therapeutic regimens.

A Hyaluronic Acid Hydrogel Loaded with Gentamicin and Vancomycin Successfully Eradicates Chronic Methicillin-Resistant Staphylococcus aureus Orthopedic Infection in a Sheep Model

Willemijn Boot et al.

Summary: The study introduces a clinically relevant large animal model of chronic MRSA ODRI, demonstrating that an injectable, thermoresponsive, hyaluronic acid-based hydrogel loaded with gentamicin and vancomycin outperforms current clinical practice treatment. These results suggest that local antibiotic delivery with an injectable hydrogel can significantly increase treatment success rates beyond current clinical practice in a robust animal model.

Bacteriophages for the Treatment of Graft Infections in Cardiovascular Medicine

Simon Junghans et al.

Summary: Bacterial infections in cardiovascular surgery pose a significant challenge, with antibiotic treatment hindered by various factors. Bacteriophage therapy, discovered a century ago, could potentially serve as an important treatment method, particularly in the face of antimicrobial resistance and personalized medicine. Further research and standardization are needed to fully utilize the potential of phage therapy in combating vascular graft infections.

Friends or Foes? Rapid Determination of Dissimilar Colistin and Ciprofloxacin Antagonism of Pseudomonas aeruginosa Phages

Katarzyna M. Danis-Wlodarczyk et al.

Summary: Research has shown that colistin significantly interferes with phage activities at its minimum inhibitory concentration, while ciprofloxacin has little impact on phages. Combination therapy with phages and antibiotics is more effective in treating P. aeruginosa, and phage replication is crucial for treatment success. The lysis-profile assays provide a useful, quick, and high-throughput method to assess antibiotic antagonism of phage infection activities.

Searching for synergy: combining systemic daptomycin treatment with localised phage therapy for the treatment of experimental pneumonia due to MRSA

Luca G. Valente et al.

Summary: The study aimed to investigate the combinatorial effects of systemic daptomycin and nebulised bacteriophages for treating experimental pneumonia caused by MRSA. Results showed that the combination of intravenous daptomycin and nebulised phages did not offer superior benefits in improving survival rates or reducing bacterial burdens in the lungs and spleen, indicating that this combination is not beneficial for patients with MRSA pneumonia.
