Co-Surfactant-Free Bioactive Protein Nanosheets for the Stabilization of Bioemulsions Enabling Adherent Cell Expansion

Alexandra Chrysanthou et al.

Summary: Bioemulsions are promising platforms for the enlargement of adherent cells and stem cells. In this study, protein nanosheets based on bovine serum albumin (BSA) and beta-lactoglobulin (BLG) were designed and functionalized with RGDSP peptides to enable cell adhesion. The results demonstrate that BLG nanosheets, even without cosurfactants, have high elasticity and can support the adhesion and proliferation of mesenchymal stem cells and human embryonic kidney (HEK) cells on bioemulsions stabilized by RGD-functionalized protein nanosheets.

Construction of Orthogonal Modular Proteinaceous Nanovaccine Delivery Vectors Based on mSA-Biotin Binding

Yixin Shi et al.

Summary: Researchers have developed a proteinaceous nanoparticle chassis that can combine with various antigens to create high-performance nanovaccines. By appending a sugar chain on the surface of the nanoparticles, they were able to increase the yield and demonstrate the ability to induce immune responses. This study provides a new technology for preparing nanoscale vaccines.

Multiplexed CuAAC Suzuki-Miyaura Labeling for Tandem Activity-Based Chemoproteomic Profiling

Jian Cao et al.

Summary: This study optimized Suzuki-Miyaura cross-coupling conditions for activity-based protein profiling and mass-spectrometry-based chemoproteomics, allowing for target deconvolution and labeling site identification. By exploiting the observed orthogonality between palladium-catalyzed cross-coupling and CuAAC, dual labeling was achieved, and the protein targets of bifunctional probes were identified through multiplexed targeted deconvolution.

Site-specific antigen-adjuvant conjugation using cell-free protein synthesis enhances antigen presentation and CD8+ T-cell response

Adam M. Weiss et al.

Summary: This study demonstrated the successful use of CFPS technology to produce large quantities of site-specific antigen-adjuvant conjugates for enhanced antigen presentation and T-cell production. Site-specific conjugation using copper-free click chemistry significantly improved the efficacy of the OVA-CpG conjugates both in vitro and in vivo, reducing the required CpG dose for T-cell production tenfold.

Production of a Promising Biosynthetic Self-Assembled Nanoconjugate Vaccine against Klebsiella Pneumoniae Serotype O2 in a General Escherichia Coli Host

Zhehui Peng et al.

Summary: A nanoscale conjugate vaccine against Klebsiella pneumoniae was successfully produced using a general Escherichia coli host system. The vaccine induced an efficient humoral immune response in draining lymph nodes and high titers of IgG antibody against bacterial lipopolysaccharide, demonstrating ideal prophylactic effects in mouse models of systemic and pulmonary infection. This study opens up possibilities for developing vaccines against other pathogenic bacteria using similar strategies.
