Rare SLC13A1 variants associate with intervertebral disc disorder highlighting role of sulfate in disc pathology

Summary: Back pain is a common and debilitating disorder with largely unknown underlying biology. In this study, a genome-wide association study was conducted using diagnoses assigned in clinical practice, and 41 variants at 33 loci were identified. The most significant association was found with a variant in the CHST3 gene, which is expressed in intervertebral discs. Rare loss-of-function variants in the SLC13A1 gene were also associated with back pain.

Genome-wide association study identifies RNF123 locus as associated with chronic widespread musculoskeletal pain

Summary: This study identified three genetic loci associated with CWP, with one locus replicated, one showing suggestive association, and one not replicated. Partial genetic correlation was found between CWP and depressive symptoms, body mass index, age of first birth and years of schooling. Tissue specificity and colocalisation analysis highlighted the relevance of skeletal muscle in CWP.
