Role of chemokines in the crosstalk between tumor and tumor-associated macrophages

Rui Qin et al.

Summary: Tumor microenvironment (TME) is a complex network consisting of various non-tumoral stromal cells, among which tumor-associated macrophages (TAMs) play a crucial role in tumor development and metastasis. Chemokines serve as important messengers in the interaction between TAMs and tumor cells. Different chemokines are involved in the generation, polarization, and suppressive function of TAMs. Modulating the effects of TAMs with chemokine antagonists shows promise as a therapeutic approach for cancer treatment.

PCSK9 promotes the progression and metastasis of colon cancer cells through regulation of EMT and PI3K/AKT signaling in tumor cells and phenotypic polarization of macrophages

Lu Wang et al.

Summary: This study found that PCSK9 plays an important role in the progression and metastasis of colon cancer by regulating tumor cell EMT and PI3K/AKT signaling, as well as the phenotypic polarization of macrophages by mediating MIF and lactate levels.

Thrombin induces IL-8/CXCL8 expression by DCLK1-dependent RhoA and YAP activation in human lung epithelial cells

Fara Silvia Yuliani et al.

Summary: Thrombin activates the DCLK1/RhoA signaling pathway, promoting YAP activation and translocation to the nucleus from the cytosol, resulting in YAP/p65 formation and binding to the NF-kappa B site, which enhances IL-8/CXCL8 expression. DCLK1 may play an essential role in thrombin-stimulated IL-8/CXCL8 expression in asthmatic lungs and suggests a potential therapeutic strategy for severe asthma treatment.

Epigenetic regulation in macrophage migration inhibitory factor (MIF)-mediated signaling in cancer and inflammation

Angelina Osipyan et al.

Summary: Epigenetic mechanisms play a crucial role in regulating tissue-specific expression of cytokine genes. MIF, a key cytokine involved in cancer and inflammation, triggers signaling pathways by binding to receptors like CD74. Abnormal expression of MIF and altered activity states of connected pathways are linked to inflammatory diseases and cancer. Therapeutic strategies based on epigenetic mechanisms have the potential to regulate MIF-mediated signaling in cancer and inflammation.

Tumour Microenvironment Stress Promotes the Development of Drug Resistance

Nicole A. Seebacher et al.

Summary: The tumour microenvironment (TME) plays a crucial role in cancer drug resistance, involving mechanisms such as altered metabolism, changes in stromal cells, increased autophagy, altered drug delivery, and induction of a cancer stem cell phenotype. There is a vast area for potential research to better understand the interplay between factors within and outside the TME, which is essential for developing new therapeutic strategies less prone to failure due to drug resistance in cancer cells.
