Tethering Piezo channels to the actin cytoskeleton for mechanogating via the cadherin-I3-catenin mechanotransduction complex

Jing Wang et al.

Summary: The mechanically activated Piezo channel is connected to the actin cytoskeleton via the cadherin-O-catenin mechanotransduction complex, enabling mechanosensitivity. Disruption of specific interactions impairs Piezo channel function.

Structure, kinetic properties and biological function of mechanosensitive Piezo channels

Xiang-Zhi Fang et al.

Summary: Mechanotransduction, Piezo channels, and their mutations play crucial roles in various biological processes, including touch sensation, balance, and cardiovascular regulation. The kinetics of Piezo channels are critical for normal physiological functions. Mutations in Piezo genes have been linked to hereditary human disorders.

Circulating prostate cancer cells have differential resistance to fluid shear stress-induced cell death

Jacob M. Hope et al.

Summary: Research shows that prostate tumor cells exhibit varying degrees of cell death response when exposed to intense fluid shear stress, with cell membrane damage, inefficient membrane repair, and increased cell stiffness playing a role in the resistance to such stress and potentially providing targets for disrupting blood-borne metastasis.

Channeling the Force: Piezo1 Mechanotransduction in Cancer Metastasis

Jenna A. Dombroski et al.

Summary: Research has shown that the Piezo1 channel plays a crucial role in cancer cell metastasis, transducing mechanical forces into biochemical signals. Calcium signaling pathways are associated with various signaling pathways related to cancer metastasis.

The fluid shear stress sensor TRPM7 regulates tumor cell intravasation

Christopher L. Yankaskas et al.

Summary: The study reveals a molecular mechanism by which cells avoid high shear during intravasation by activating TRPM7 channel to promote extracellular calcium influx, which then activates different pathways to coordinate the reversal of migration direction and avoid shear stress. Cells with higher TRPM7 activity levels intravasate less efficiently and establish less invasive metastatic lesions. This study provides a mechanistic interpretation for the role of shear stress and its sensor, TRPM7, in tumor cell intravasation.

The migration of metastatic breast cancer cells is regulated by matrix stiffness via YAP signalling

Wei Chen et al.

Summary: Matrix stiffness influences the proliferation and migration ability of breast cancer cells, with YAP playing a significant role in this process. As matrix stiffness increases, highly metastatic breast cancer cells exhibit a different migratory potential compared to non-metastatic cells, and this process is YAP-dependent.
