Helicobacter pylori-induced reactive oxygen species direct turnover of CSN-associated STAMBPL1 and augment apoptotic cell death

Supattra Chaithongyot et al.

Summary: STAMBPL1, a deubiquitinylase, plays an important role in cell survival and regulating cell death. This study found that chemotherapeutic agents and Helicobacter pylori can induce the degradation of STAMBPL1, and its interaction with CSN is crucial for its stability and function. Additionally, STAMBPL1 can deubiquitinylate the anti-apoptotic protein Survivin, enhancing cell survival.

Co-adaptor driven assembly of a CUL3 E3 ligase complex

David Akopian et al.

Summary: Cuilin-RING E3 ligases (CRLs) are essential ubiquitylation enzymes and the formation of CRL complexes is a tightly regulated process, with co-adaptors playing an important role in CRL assembly.

Deciphering the LRRK code: LRRK1 and LRRK2 phosphorylate distinct Rab proteins and are regulated by diverse mechanisms

Asad U. Malik et al.

Summary: Autosomal dominant mutations in LRRK2 enhance kinase activity and cause Parkinson's disease. LRRK2 phosphorylates Rab GTPases including Rab8A and Rab10. This study found that LRRK1 can phosphorylate Rab7A at Ser72, and identified mutations that enhance this phosphorylation.

CSN7B defines a variant COP9 signalosome complex with distinct function in DNA damage response

Jing Wang et al.

Summary: CSN7A and CSN7B have overlapping functions in deneddylation, but CSN7B plays a unique role in DNA damage sensing and affects DNA repair pathway choice; knockout of CSN7B leads to resistance to DNA damage and a switch in DNA repair pathway.

IP6-assisted CSN-COP1 competition regulates a CRL4-ETV5 proteolytic checkpoint to safeguard glucose-induced insulin secretion

Hong Lin et al.

Summary: The study uncovers an antagonistic relationship between COP1 and CSN in glucose-induced insulin secretion, with disruption of the CRL4(COP1)-ETV5 proteolytic axis leading to hyperinsulinemia and obesity. The findings suggest that deregulation of the IP6-CSN-CRL4(COP1)-ETV5 axis underlies these pathologies, and that targeting this pathway could reduce the risk of obesity and diabetes. Furthermore, inhibition of CRL neddylation can correct these pathological phenotypes.
