Oxidative Stress and Lipid Accumulation Augments Cell Death in LDLR-Deficient RPE Cells and Ldlr -/- Mice

Parameswaran Gangadharan Sreekumar et al.

Summary: Lipid peroxidation caused by oxidative stress is a major factor in age-related macular degeneration (AMD). This study aims to investigate the role of LDLR in the NaIO3-induced model of dry AMD. The results indicate that LDLR loss leads to lipid accumulation and impaired retinal function, which may contribute to the development of AMD.

Retinal cholesterol metabolism is perturbated in response to experimental glaucoma in the rat

Elise Leger-Charnay et al.

Summary: This study reveals that ocular hypertension is associated with significant changes in retinal cholesterol metabolism. Genes involved in cholesterol biosynthesis and uptake were upregulated, while genes involved in efflux were downregulated. This dysregulation led to increased cholesterol levels, but counter-regulatory mechanisms were activated to restore cholesterol homeostasis and promote ganglion cell viability.

Omega-3/Omega-6 Long-Chain Fatty Acid Imbalance in Phase I Retinopathy of Prematurity

Zhongjie Fu et al.

Summary: There is a gap in understanding the effect of omega-3 and omega-6 long-chain polyunsaturated fatty acids (LCPUFA) on Phase I retinopathy of prematurity (ROP). This study found that dietary omega-3 supplementation promoted retinal vessel development in mouse neonates with hyperglycemia-associated Phase I retinopathy. However, omega-6 LCPUFA was also essential for neuronal growth and metabolism. The activation of the adiponectin (APN) pathway by omega-3 LCPUFA protected against retinal neurovascular dysfunction in ROP.

Accumulation of Lipid Droplets in a Novel Bietti Crystalline Dystrophy Zebrafish Model With Impaired PPAR? Pathway

Pan Gao et al.

Summary: A zebrafish model was generated to study the functional role of CYP4V2 in BCD development, revealing abnormal lipid metabolism in RPE cells as a direct consequence of CYP4V2 deficiency. Transcriptome analysis suggested dysregulation of the PPAR alpha pathway as a potential therapeutic target for BCD.

Lipid metabolism dysregulation in diabetic retinopathy

Julia Busik

Summary: This review article discusses the potential benefits of lipid-lowering drugs for the prevention and treatment of diabetic retinopathy. While fibrates have shown improvement in DR, there is a disconnect between their effects in the retina and blood lipid levels. The article also highlights lipid and lipoprotein-independent mechanisms that may play a role in retinal protection.

Astrocytes in stress accumulate lipid droplets

Tina Smolic et al.

Summary: The study found that the increase of LDs in astrocytes may be crucial for cell survival and proliferation. Under stress conditions, exposure to noradrenaline and metabolic stress strongly facilitated LD accumulation in astrocytes, potentially providing energy support and neuroprotection against stress-induced lipotoxicity.

Retinal bioavailability and functional effects of a synthetic very-long-chain polyunsaturated fatty acid in mice

Aruna Gorusupudi et al.

Summary: The study synthesized a sufficient amount of very-long-chain polyunsaturated fatty acid (32:6 n-3) for research in the mouse retina, showing that VLC-PUFA-fed mice had significantly increased levels of retinal VLC-PUFA and improvements in visual acuity and electroretinography measurements compared to controls. This suggests potential benefits of synthetic VLC-PUFAs for the treatment and prevention of retinal degenerative diseases.

Metabolism in Retinopathy of Prematurity

Yohei Tomita et al.

Summary: Retinopathy of prematurity is a condition where retinal abnormalities occur in premature infants due to disturbed oxygen conditions and nutrient supply. Current therapeutic interventions target hypoxia-induced neovessels but can have negative impacts. Metabolic disturbance, such as hyperglycemia and dyslipidemia, is an underexplored risk factor in the disease pathogenesis.
