Bcl-xL acts as an inhibitor of IP3R channels, thereby antagonizing Ca2+-driven apoptosis

Nicolas Rosa et al.

Summary: The study shows that Bcl-xL and Bcl-2 have similar anti-apoptotic effects in controlling endoplasmic reticulum Ca2+ release, challenging the current understanding of their divergent functions in Ca2+ signaling modulation.

DRP1 interacts directly with BAX to induce its activation and apoptosis

Andreas Jenner et al.

Summary: BAX and DRP1 physically interact and their interaction is enhanced during apoptosis. This interaction occurs exclusively in the membrane environment and involves different surfaces of BAX. Additionally, the association between BAX and DRP1 enhances their membrane activity. Forced dimerization of BAX and DRP1 triggers their activation and translocation to mitochondria, inducing mitochondrial remodeling and permeabilization, leading to apoptosis even in the absence of apoptotic triggers. Therefore, it is proposed that DRP1 can promote apoptosis by acting as a noncanonical direct activator of BAX through physical contacts with its N-terminal region.

The BAD-BAX-Caspase-3 Cascade Modulates Synaptic Vesicle Pools via Autophagy

Qinhua Gu et al.

Summary: The BAD-BAX-caspase-3 cascade and autophagy play new roles in controlling synaptic vesicle pools, influencing the kinetics of synaptic vesicle pool activity-induced depletion and recovery, and facilitating associative fear learning and memory. This study uncovers a novel mechanism for controlling synaptic vesicle pools and a nonapoptotic presynaptic function of the BAD-BAX-caspase-3 cascade.

Bcl-2 Family of Proteins in the Control of Mitochondrial Calcium Signalling: An Old Chap with New Roles

Summary: Bcl-2 family proteins are not only important regulators of apoptosis, but are also emerging as regulators of intracellular calcium concentrations. They interact with major calcium transporters at mitochondria-ER contact points, controlling mitochondrial calcium balance and impacting cell survival and migration.
