Summary: Microbial conversion of waste glycerol into value-added chemicals, such as 3-hydroxypropionic acid (3-HP), is an important bioprocessing technology. However, commercial exploitation of 3-HP biosynthesis faces challenges including toxic effects, undesirable carbon flux, low tolerance, enzyme co-factor dependence, low enzyme activity and stability, substrate inhibition and specificity. This review critically discusses the fundamentals, challenges, recent research efforts, and outlines possible strategies for sustainable and industrially exploitable 3-HP production.

Benchmarking recombinant Pichia pastoris for 3-hydroxypropionic acid production from glycerol

Summary: This study demonstrated the use of P. pastoris as a cell factory to produce 3-hydroxypropionic acid from glycerol. By introducing the mcr gene from Chloroflexus aurantiacus and optimizing enzyme and metabolic engineering modifications, a significant increase in 3-HP production was achieved. The best strain (PpHP6) showed a high volumetric productivity, indicating the promising potential of P. pastoris for the production of bulk chemicals, especially 3-HP, from crude glycerol.
