Summary: This study suggests that CB2 activation may alleviate HIV-associated antiretroviral neuropathy and identifies a previously unreported mechanism through which CB2 activation produces antinociceptive efficacy. The study also demonstrates that a CB2 agonist can reverse established morphine tolerance and shows that CB2 localized to peripheral sensory neurons mediates the opioid tolerance sparing efficacy of CB2 agonists.

Summary: There are inherent structural and functional differences in the central nervous systems of females and males, including sex-specific differences in genetic, epigenetic, and hormonal factors. Microglia, as principal resident macrophages in the CNS, have recently been found to exhibit sex-dependent structural and functional differences, which has implications for the design of sex- and microglia-specific therapeutic approaches for CNS diseases.

Spared Nerve Injury Causes Sexually Dimorphic Mechanical Allodynia and Differential Gene Expression in Spinal Cords and Dorsal Root Ganglia in Rats

Summary: Neuropathic pain is more prevalent in women, but the mechanisms of sex differences are not fully understood. Using a rat model, it was found that female rats exhibited stronger mechanical allodynia, and numerous sexually dimorphic changes in gene expression were observed following SNI. These changes involved genes related to T-cells, immunological responses, neuronal transmission, and plasticity, suggesting potential gene targets for sex-specific therapies in neuropathic pain.

Activation of Neuroprotective Microglia and Astrocytes at the Lesion Site and in the Adjacent Segments Is Crucial for Spontaneous Locomotor Recovery after Spinal Cord Injury

Summary: The study demonstrates that the first week post-injury is critical for the modulation of reactive microglia and astrocytes into their neuroprotective phenotypes. Gene expression data showed a significant upregulation of M1 microglia and astrocytes at the lesion site one week after spinal cord injury, which gradually attenuated after two weeks post-injury. Furthermore, a positive correlation was observed between neurological outcomes and the expression of certain anti-inflammatory genes, indicating the importance of early intervention in promoting neuroprotective phenotypes of glial cells after injury.
