The Role of Macrophages in the Pathogenesis of Atherosclerosis

Summary: This review summarizes the involvement of various cell populations, including immune cells and endothelial cells, in the pathogenesis of atherosclerosis. Among them, macrophages are of particular importance due to their diverse effects on atherogenesis and inflammation in atherosclerosis. The significance of different phenotypes of macrophages in the progression or regression of atherosclerosis is discussed, including their ability to become foam cells, the consequences of this event, and their ability to create a pro-inflammatory or anti-inflammatory environment through cytokine production at the site of atherosclerotic lesions. In addition, therapeutic strategies aimed at modulating macrophage activity are considered, providing useful ideas for future drug developments.

Pro-inflammatory role of Wnt/β-catenin signaling in endothelial dysfunction

Summary: In this study, the potential of a Wnt/beta-catenin signaling inhibitor, iCRT-14, in reversing the adverse effects of TNF-alpha on endothelial physiology was evaluated. The results showed that iCRT-14 could decrease inflammatory cytokine production, monocyte adhesion, and endothelial permeability, and increase thrombosis. However, iCRT-14 also exerted a moderate inhibitory effect on wound healing, which may affect its suitability as a therapy for atherosclerosis and vein graft failure.

Shear stress control of vascular leaks and atheromas through Tie2 activation by VE-PTP sequestration

Summary: Vascular endothelial protein tyrosine phosphatase (VE-PTP) affects endothelial barrier function by regulating Tie2 activation. High shear stress caused VE-PTP polarization and endocytosis, along with Tie2 activation, in regions with smooth flow. In regions with disturbed flow, VE-PTP remained associated with Tie2. Endothelial cells exposed to high shear stress had increased Tie2 activation and reduced macromolecular permeability. Deleting VE-PTP or inhibiting it with AKB-9785 had similar anti-atherogenic effects.

The taxonomy mapping enables analysis of the conservation of functional and physical links among genes in different organism groups.

Participation of Kruppel-like Factors in Atherogenesis

Summary: Atherosclerosis is a significant issue in modern medicine, with many aspects not fully understood by clinicians. It develops through a complex chain of events involving various cells of the vascular wall and peripheral blood flow. Endothelial cells, as the lining of blood vessels, have a crucial role in vascular biology, not only acting as a barrier between blood flow and tissues but also regulating hemodynamics and influencing other cells in the vascular wall. Kruppel-like factors (KLFs) have diverse biological functions in different cells of the vascular wall, particularly KLF2 and KLF4, which play important roles in both normal vascular function and atherosclerosis.

Effects of shear stress on vascular endothelial functions in atherosclerosis and potential therapeutic approaches

Summary: Different blood flow patterns in arteries alter the phenotype of vascular endothelial cells, affecting their functions and directly contributing to the occurrence of early atherosclerosis lesions. Oscillating shear stress (OSS) in curved or bifurcated arteries promotes the transformation of endothelial cells into pro-inflammatory phenotypes, leading to increased inflammation, oxidative stress, mitochondrial dysfunction, metabolic abnormalities, and enhanced endothelial permeability, thus promoting atherosclerosis progression. In contrast, stable laminar shear stress (LSS) in straight arteries facilitates the transformation of endothelial cells into an anti-inflammatory phenotype, improving their function and inhibiting atherosclerotic progression. This article outlines the relationship between vascular endothelial cell functions and different forms of fluid shear stress in atherosclerosis and explores potential therapeutic approaches targeting intracellular mechanical transductions.
