Summary: This article discusses the impact of Alzheimer's disease on public health, including its prevalence and incidence, mortality and morbidity, care usage and costs, and the effects on family caregivers, the dementia workforce, and society. It highlights the patient journey and potential treatment options for Alzheimer's disease. Currently, there are 6.7 million older Americans living with Alzheimer's dementia, a number that could increase to 13.8 million by 2060 without medical breakthroughs. Alzheimer's is the sixth-leading cause of death in the US, with deaths increasing by over 145% between 2000 and 2019. There is a decline in the number of caregivers and an increase in care provided by each remaining caregiver. The costs of unpaid caregiving in 2022 reached $339.5 billion, impacting caregivers' mental and physical health. Additionally, there is a shortage of healthcare workers in dementia care, and Medicare and Medicaid payments are significantly higher for individuals with dementia. The article also explores the need for sufficient physician specialists in Alzheimer's care, considering the availability of drugs that change the underlying biology of the disease.

Hippocampus and its involvement in Alzheimer's disease: a review

Y. Lakshmisha Rao et al.

Summary: The hippocampus is a crucial component of the brain involved in learning and memory. Alzheimer's disease can lead to tissue loss in the hippocampus and impair its function. External factors such as diet and physical activity can influence neural stem cell function and delay neurodegeneration.

Age-dependent formation of TMEM106B amyloid filaments in human brains

Manuel Schweighauser et al.

Summary: The study found that TMEM106B protein forms amyloid filaments in the brains of patients with various neurodegenerative diseases. The structures of TMEM106B filaments differ in different brain regions and do not show clear associations with specific diseases. The presence of TMEM106B filaments in the brains of older individuals suggests an age-dependent formation pattern, which is not observed in younger individuals with normal neurology.

Waste Clearance in the Brain and Neuroinflammation: A Novel Perspective on Biomarker and Drug Target Discovery in Alzheimer's Disease

Kazuhiko Uchida

Summary: Alzheimer's disease is a complex disease with a heterogeneous etiology. Recent research has shown that neuroinflammation plays a significant role in the pathology of Alzheimer's disease. Dysfunction in waste clearance may serve as a biomarker for early cognitive impairment. Improving waste clearance could be an early intervention strategy for Alzheimer's disease and preclinical stages of dementia.

Dementia Epidemiology Fact Sheet 2022

Joon-Ho Shin

Summary: This fact sheet provides an overview of dementia epidemiology in Korea, using representative government-led data. It summarizes the prevalence and incidence of the condition using various types of data. Dementia prevalence and incidence have been increasing and are predicted to continue doing so. This information will be used by public health officials, healthcare professionals, and policymakers to develop strategies for dementia rehabilitation and prevention.

Summary: This article discusses the impact of Alzheimer's disease on public health, including its incidence, prevalence, mortality and morbidity rates, healthcare costs and caregivers' burden. It also addresses the perspectives of consumers and primary care physicians on mild cognitive impairment (MCI) and the need for improved awareness, diagnosis, and treatment. The article emphasizes the increasing number of individuals living with Alzheimer's and the challenges faced by caregivers and healthcare providers, particularly during the COVID-19 pandemic.

Familial Alzheimer's disease mutations in amyloid protein precursor alter proteolysis by γ-secretase to increase amyloid β-peptides of ≥45 residues

Sujan Devkota et al.

Summary: Systematic and quantitative analyses were conducted to study the effects of 14 FAD mutations on γ-secretase processing of APP substrates. All mutations resulted in inefficient processing of Aβ peptides longer than 45 residues. Certain mutations provided insights into the mechanism of processive proteolysis.

Summary: Alzheimer's disease is a chronic neurodegenerative disease related to the overproduction and accumulation of amyloid-beta peptide in the brain, for which there is currently no disease-modifying therapy available. Recent research suggests a possible infectious cause for AD, with multiple pathogens playing a potential role in its pathogenesis.
