Evaluating the field performance of multiple SARS-Cov-2 antigen rapid tests using nasopharyngeal swab samples

Magyar Nora et al.

Summary: The SARS-CoV-2 pandemic has posed significant challenges to the global health care system. Early diagnosis is crucial to limit the spread of the virus, but factors such as availability of laboratory equipment and trained personnel affect the use of time-consuming molecular techniques. To facilitate on-the-spot diagnosis, several SARS-CoV-2 rapid antigen tests have been developed. A study in Hungary evaluated the performance of ten commercially available rapid tests compared to RT-qPCR. The results showed promising sensitivity and specificity for these rapid antigen tests.

Verification of lateral flow antigen tests for SARS-CoV-2 by qPCR directly from the test device

Ludwig Czibere et al.

Summary: This study aimed to combine the speed of lateral flow antigen tests (LFATs) with the reliability of qPCR tests for SARS-CoV-2 detection. The results showed that nucleic acid amplification systems had higher sensitivity compared to LFAT. Amplification using extracted eluates improved sensitivity by 10-100 times. LFAT is recommended for single samples in emergency clinical or dental settings, as well as for smaller and larger population screening due to its low cost and fast detection.

Mutations in SARS-CoV-2 nucleocapsid in variants of concern impair the sensitivity of SARS-CoV-2 detection by rapid antigen tests

Ibrahim T. Hagag et al.

Summary: Rapid antigen tests (RATs) are convenient tools for SARS-CoV-2 testing, but they have lower sensitivity and may misdiagnose samples with low viral loads. This study examined the analytical sensitivities of different RATs and found that certain amino acid substitutions can affect their sensitivity. Therefore, optimizing the antibodies used in lateral flow assays is necessary.
