Brain injury pathophysiology study by a multimodal approach in children with sickle cell anemia with no intra or extra cranial arteriopathy

Valentine Brousse et al.

Summary: This study aims to explore the pathophysiology and treatment methods of silent cerebral infarcts (SCI) in children with sickle cell anemia (SCA). The study found that children with SCI have decreased blood pressure, impaired blood rheology, increased P-selectin levels, and marked anemia. ASL may serve as an additional screening tool, and early treatment can reduce the risk of this complication.

Bone loss is ameliorated by fecal microbiota transplantation through SCFA/GPR41/ IGF1 pathway in sickle cell disease mice

Liping Xiao et al.

Summary: The study suggests that transferring fecal microbiota from healthy control mice to SCD mice can significantly improve bone loss, enhance the expression of bone formation genes, increase SCFA levels, and partially restore IGF1 levels, thereby protecting SCD patients from bone loss.

Insulin-like growth factor binding protein-1 regulates HIF-1α degradation to inhibit apoptosis in hypoxic cardiomyocytes

Xiaoyan Tang et al.

Summary: This study demonstrated that hypoxia up-regulates IGFBP-1 and HIF-1 alpha expression in cardiomyocytes. Additionally, IGFBP-1 regulates HIF-1 alpha stabilization through interacting with VHL. Therefore, IGFBP-1-HIF-1 alpha could be targeted for treating ischemic heart disease.

MRI detection of brain abnormality in sickle cell disease

Hanne Stotesbury et al.

Summary: Recent studies have shown that quantitative MRI techniques can identify structural and hemodynamic biomarkers associated with the risk of neurological and neurocognitive complications in patients with sickle cell disease. Evidence suggests that these biomarkers are sensitive to changes with treatments like blood transfusion and hydroxyurea, making them potential endpoints in future clinical trials of novel approaches. With further validation, these techniques may also improve risk stratification for neurological and neurocognitive complications in this vulnerable population.

Preclinical and clinical evidence of IGF-1 as a prognostic marker and acute intervention with ischemic stroke

Cellas A. Hayes et al.

Summary: Reduced levels of IGF-1 are associated with increased mortality, poorer functional outcomes, and worsened morbidity following ischemic stroke. Administering exogenous IGF-1 before or after ischemic insult can attenuate neurological damage and behavioral changes. Research reveals the significant role of IGF-1 in the process of stroke damage.

Hypoxia Inducible Factor-1α Attenuates Ischemic Brain Damage by Modulating Inflammatory Response and Glial Activity

Nashwa Amin et al.

Summary: The study found that in a focal cerebral ischemia mouse model, the activator of hypoxia-inducible factor 1 significantly increased IL-10 levels while decreasing inflammatory factors iNOS, TNF-alpha, and NF-kB levels, exerting an anti-inflammatory role to alleviate brain damage. Additionally, astrocytic activation was inhibited, and hypoxia-inducible factor 1 induction alleviated neuronal soma damage and cell death.
