DNA Damage Response Mechanisms in Head and Neck Cancer: Significant Implications for Therapy and Survival

Summary: Head and neck cancer (HNC) is a heterogeneous group of tumors that commonly occur in the oral cavity, larynx, nasopharynx, oropharynx, and hypopharynx. Despite advancements in treatment, HNC has a high recurrence rate and poor prognosis for metastatic patients. HNCs are characterized by genomic instability and defective DNA damage repair processes, which play a role in response to treatment and potential development of new therapies. This review summarizes the understanding of deregulated genes and proteins involved in DNA damage repair pathways in HNC, their implications in disease development, and the potential for targeted therapies.

Incidence and mortality trends of nasopharynx cancer from 1990 to 2019 in China: an age-period-cohort analysis

Summary: This study analyzed the long-term trends of nasopharynx cancer (NPC) incidence and mortality in China. The findings showed an increase in incidence rate but a decrease in mortality rate over the past three decades. The study also highlighted the age and gender differences in NPC incidence. The global, regional, and national burden of nasopharyngeal carcinoma and its attributable risk factors in 204 countries and territories, 1990-2019

Summary: This study provides an overview of the global epidemiology of nasopharyngeal cancer (NPC) and reveals that the highest incidence rates are in East Asia. From 1990 to 2019, the incidence rates in East Asia and High-income Asia Pacific had the greatest increase, while Central Asia and the Caribbean experienced the greatest increase in disability-adjusted life-years (DALY) and death rates. At the national level, Cabo Verde, Romania, and Cyprus had the largest increases in incidence rates, while Cabo Verde, Romania, and Jamaica had the largest increases in DALY and death rates.

Multi-pathway DNA-repair reporters reveal competition between end-joining, single-strand annealing and homologous recombination at Cas9-induced DNA double-strand breaks

This study developed fluorescent Cas9-based reporters to quantify the contribution of multiple DNA repair pathways. LncRNA linc00312 suppresses radiotherapy resistance by targeting DNA-PKcs and impairing DNA damage repair in nasopharyngeal carcinoma

Summary: Our study found that linc00312 is significantly down-regulated in NPC tissues and higher expression of linc00312 is associated with longer overall survival and better short-term radiotherapy efficacy. Overexpression of linc00312 increases NPC cells' sensitivity to ionizing radiation by hindering the DNA repair and cell cycle control mechanisms. DNA-PKcs was identified as the binding protein of linc00312, revealing a novel mechanism in the DNA damage response.
