Pluripotent stem cell-derived committed cardiac progenitors remuscularize damaged ischemic hearts and improve their function in pigs

In Vitro Matured Human Pluripotent Stem Cell-Derived Cardiomyocytes Form Grafts With Enhanced Structure and Function in Injured Hearts

Summary: The study showed that culturing hPSC-CMs on PDMS-lined roller bottles and transplanting these cells into a guinea pig model of myocardial infarction resulted in better structural and functional outcomes compared to conventional immature hPSC-CM populations. The PDMS-matured hPSC-CMs exhibited increased cardiac gene expression and more mature properties in vitro, and the intracardiac grafts formed with these cells showed enhanced structure and alignment, better electromechanical integration, and improved contractile function. This study provides practical methods for the scaled generation of mature hPSC-CMs and highlights the importance of using more mature cardiomyocytes for cardiac regeneration.

Pharmacologic treatment of EA may be a viable strategy to improve safety and allow further clinical development of cardiac remuscularization therapy.
