Association between IGF-1 levels ranges and all-cause mortality: A meta-analysis

Summary: The association between IGF-1 levels and all-cause mortality is complex, with both low and high levels being associated with increased mortality risk. This meta-analysis found that higher IGF-1 levels were not associated with increased all-cause mortality compared to low levels. However, there was a U-shaped relationship between IGF-1 levels and mortality risk, with both low and high IGF-1 levels increasing the risk compared to middle levels. A specific range of 120-160 ng/ml was associated with the lowest mortality risk.

Progerin modulates the IGF-1R/Akt signaling involved in aging

Summary: Progerin, a result of LMNA mutation, causes nuclear abnormalities and premature aging. Impaired IGF-1R/Akt signaling pathway leads to growth retardation and aging. Progerin mislocalizes outside of nucleus, interacts with IGF-1R, and down-regulates its expression, resulting in inhibited mitochondrial respiration, retarded cell growth, and accelerated senescence. PTEN inhibitor bpV (HOpic) increases Akt activity and improves abnormalities in progeria. These findings provide important insights into the role of Progerin in regulating IGF-1R/Akt signaling and have potential implications in treating LMNA-associated progeroid disorders.

Summary: As individuals age, the function of hematopoietic stem cells (HSCs) declines, leading to limited health span of the blood and immune systems. Research indicates that the hallmarks of aging in HSCs and hematopoiesis start accumulating by middle age, with decreased levels of the longevity-associated molecule IGF1 in the bone marrow microenvironment playing a key role in causing HSC aging. By directly stimulating middle-aged HSCs with IGF1, molecular and functional hallmarks of aging can be rescued, highlighting the complex relationship between IGF1 levels, longevity, and HSC function.
