Identification of Molecular Determinants in iRhoms1 and 2 That Contribute to the Substrate Selectivity of Stimulated ADAM17

Yi Zhao et al.

Summary: The functional properties of ADAM17/iRhom1 and ADAM17/iRhom2 complexes differ, with iRhom2 supporting more stimulated shedding of substrates compared to iRhom1. The TMD7 of the iRhoms plays an important role in determining substrate selectivity.

Analysis of the Conditions That Affect the Selective Processing of Endogenous Notch1 by ADAM10 and ADAM17

Rolake O. Alabi et al.

Summary: The Notch signaling pathway relies on the roles of proteins like ADAM10 and ADAM17 to regulate cell fate, with ADAM17 preferring to process Notch1 in ligand-independent conditions, and ADAM10 preferring to process Notch1 in ligand-dependent conditions. These findings provide new insights into the substrate selectivity mechanisms of ADAM10 and ADAM17 towards Notch1.

Members of the Fibroblast Growth Factor Receptor Superfamily Are Proteolytically Cleaved by Two Differently Activated Metalloproteases

Summary: Metalloproteases ADAM10 and ADAM17 have been identified as differentially regulated FGFR membrane sheddases, playing roles in modulating FGFR signaling during cell migration. Ectodomain shedding can impact the function of ligand-induced FGFR signaling in cell movement.

Summary: Enzymes are no longer considered static molecular machines, as they can move to different locations and change their functions. Recent research focuses on the regulation and function of ADAM endopeptidases at specific subcellular sites or in multimolecular complexes, particularly ADAM10 and TACE/ADAM17.

ADAM17-A Potential Blood-Based Biomarker for Detection of Early-Stage Ovarian Cancer

Christoph Rogmans et al.

Summary: Ovarian cancer is characterized by high mortality rates due to late diagnosis and lack of early symptoms. ADAM17, a metalloprotease, shows promise as a potential diagnostic blood-based biomarker in ovarian cancer, as it is highly expressed in various solid tumors. This study found that ADAM17 was detected in ovarian cancer cell lines, primary cells, serum, and ascites of patients, with significantly higher levels in ovarian cancer patients compared to controls. Moreover, ADAM17 levels were elevated in early stages of the disease, suggesting its potential use in combination with other markers for early detection of ovarian cancer.
