Brome mosaic virus detected in Kansas wheat co-infected with other common wheat viruses

Nar B. Ranabhat et al.

Summary: Wheat breeders are using new sequencing technology to study the natural field virus populations in Kansas. They found brome mosaic virus (BMV) present in 29 out of 47 sampled counties, often co-infected with other wheat viruses. The study also revealed the genetic characteristics and variations of BMV isolates. This information is important for plant breeding and improving monitoring tools for virus detection.

Molecular Approaches for Detection of Trichoderma Green Mold Disease in Edible Mushroom Production

Ljiljana Sasic Zoric et al.

Summary: The green mold disease caused by Trichoderma spp. is highly detrimental to edible mushroom production. This review discusses the molecular methods used for detection of green mold disease, including nucleic acid-based methods and isothermal amplification methodology for field applications. Early detection of Trichoderma spp. is crucial for prevention and control measures due to the aggressive nature of the disease and its economic impact. Challenges and future perspectives in Trichoderma diagnostics, as well as fast screening molecular methods, are also discussed.

Identification of Viruses Infecting Oats in Korea by Metatranscriptomics

Na-Kyeong Kim et al.

Summary: This study identified three different virus species infecting oats in six different geographical locations in Korea using RNA-sequencing and bioinformatics analyses. The dominant virus infecting winter oats was barley yellow dwarf virus (BYDV), while rice black-streaked dwarf virus (RBSDV) was identified in only one region. The genetic variations and phylogenetic analyses provided insights into the characteristics and origins of these viruses.

Wheat cultivars and natural-based substances: Impacts on epidemiological parameters of yellow dwarf disease

Elodie Pichon et al.

Summary: The control methods against aphid-transmitted B/CYDV in cereal fields have mainly relied on neonicotinoid-coated seeds, but the ban of neonicotinoid by the EU has sparked interest in genetic resources and biological protections for alternative management strategies. This study evaluated the potential of 11 susceptible wheat varieties to affect aphid biology and virus spread, and tested natural-based substances for their impact on aphid survival, colonization, and virus transmission. The results showed that some wheat cultivars exhibited partial resistance phenotypes and that azadirachtin increased aphid mortality and decreased fecundity. Thus, future management strategies against yellow dwarf disease should consider the genetic backgrounds of susceptible wheat cultivars and the use of azadirachtin.

Current Developments and Challenges in Plant Viral Diagnostics: A Systematic Review

Gajanan T. Mehetre et al.

Summary: Plant viral diseases pose a significant threat to sustainable agriculture, and timely and accurate identification of causative agents is crucial for effective treatment. Current diagnostic methods primarily involve serological and nucleic acid techniques, but there is a growing interest in developing real-time diagnostic tools to improve in-field diagnosis capabilities. Next-generation sequencing (NGS) shows great potential for detecting multiple viruses simultaneously, but further research is needed to optimize these techniques for plant virology.

Molecular characterization of a novel wheat-infecting virus of the family Betaflexiviridae

Shuai Fu et al.

Summary: Plants showing symptoms of yellowing, mosaic, and stunting were collected from wheat fields in Henan Province, China. Analysis revealed a novel virus, tentatively named wheat yellow stunt-associated betaflexivirus (WYSaBV), which is evolutionarily related to the subfamily Quinvirinae within the family Betaflexiviridae. This virus contains six open reading frames and distinct features from other established genera.

Application of Oxford Nanopore Technology to Plant Virus Detection

Lia W. Liefting et al.

Summary: The adoption of Oxford Nanopore Technologies (ONT) sequencing in plant virology has been slow, but its promise of real-time generation of long nucleotide sequences makes it an attractive method for both low- and high-scale virus diagnostics. The portability of the MinION and Flongle platforms, combined with lower costs and improved read accuracy, highlight ONT's feasibility as a valuable component to diagnostician's toolkit.
