Screening of Monoamine Oxidase B Inhibitors from Fragaria nubicola by Ligand Fishing and Their Neuroprotective Effects

Summary: In this study, MAO-B inhibitors in Tibetan strawberry were screened using enzyme-functionalized magnetic nanoparticles coupled with UPLC-QTOF-MS. Two inhibitors, quercetin-3-O-beta-D-glucuronide-6''-methyl ester (1) and kaempferol-3-O-beta-D-glucuronide-6''-methyl ester (2), were identified with IC50 values of 19.44 +/- 1.17 and 22.63 +/- 1.78 mu M, respectively. The compounds also exhibited significant neuroprotective effects on PC12 cells.

Summary: This study aimed to screen and identify MAO-B inhibitors from the roots of Platycodon grandiflorus using MAO-B functionalised MNPs, HPLC-MS, and NMR analysis. Two inhibitors, protocatechuic aldehyde and coumarin, were identified with potential anti-PD activity, especially protocatechuic aldehyde that also increased cell viability. The results shed light on the anti-PD activity of Platycodon grandiflorus roots and the efficacy of using ligand fishing method for discovering MAO-B inhibitors.

Effects of Natural Monoamine Oxidase Inhibitors on Anxiety-Like Behavior in Zebrafish

Summary: In this study, the effects of natural MAO inhibitors on anxiety behavior induced by novel environment in zebrafish were investigated. Harmane, norharmane, and TIQ were found to have anxiolytic-like effects, while TMN showed a mix of anxiolytic- and anxiogenic-like effects. The brain bioavailability of these compounds was high, suggesting that the observed effects on anxiety-like behavior in zebrafish were likely due to their direct effects in the brain.
