Adverse effects of polymyxin B administration to healthy horses

Julia N. van Spijk et al.

Summary: PolyB treatment in horses can cause transient ataxia, which worsens with cumulative doses of PolyB and co-administration of gentamicin. Nephrotoxicity of PolyB is only evident when combined with gentamicin.

Omeprazole Prevents Colistin-Induced Nephrotoxicity in Rats: Emphasis on Oxidative Stress, Inflammation, Apoptosis and Colistin Accumulation in Kidneys

Mohammed Z. Nasrullah et al.

Summary: The concomitant administration of omeprazole significantly reduces colistin-induced nephrotoxicity in rats by preventing abnormal elevation of renal biomarkers, reducing histopathological changes, and inhibiting oxidative stress and inflammation. Omeprazole also prevents the toxic accumulation of colistin in kidneys.

Population pharmacokinetic and optimization of polymyxin B dosing in adult patients with various renal functions

Summary: Renal function significantly impacts the pharmacokinetics of polymyxin B, and dosing of polymyxin B should be adjusted according to CrCL in patients with renal insufficiency.

Systematic review on estimated rates of nephrotoxicity and neurotoxicity in patients treated with polymyxins

Florian Wagenlehner et al.

Summary: Polymyxin treatment is associated with a higher risk of nephrotoxicity compared to non-polymyxin-based regimens. The nephrotoxicity rate is higher in patients receiving polymyxins, while there is no significant difference in neurotoxicity between polymyxin and non-polymyxin treatments.

Gentamicin plasma concentrations in hospitalized horses and retrospective minimal inhibitory concentrations of gram-negative equine pathogens

Angelika Schoster et al.

Summary: The optimal dosage regimen of gentamicin in horses is still being investigated. Administering 10 mg/kg of gentamicin to hospitalized horses can reach target plasma concentrations in most cases. Factors like therapeutic versus prophylactic administration, primary disease, azotemia, and systemic inflammatory response syndrome do not affect the achievement of desired peak or trough levels.

Comparative Effects of Metamizole (Dipyrone) and Naproxen on Renal Function and Prostacyclin Synthesis in Salt-Depleted Healthy Subjects-A Randomized Controlled Parallel Group Study

Lea S. Blaser et al.

Summary: The study found that short-term use of metamizole or naproxen has no significant effect on renal function in healthy subjects on a low sodium diet, but reduces the renal excretion of 6-keto-prostaglandin F1 alpha. Pharmacokinetic analysis showed that at clinically effective doses, both 4-MAA and naproxen inhibit COX-mediated renal prostacyclin synthesis.
