Production of a Series of Long-Chain Isomaltooligosaccharides from Maltose by Bacillus subtilis AP-1 and Associated Prebiotic Properties

Molecular Docking and Site-Directed Mutagenesis of GH49 Family Dextranase for the Preparation of High-Degree Polymerization Isomaltooligosaccharide

Summary: In this study, we used molecular docking technique to study the potential effect of key amino acids in the engineered strain expressed dextranase on its enzymatic properties. The results showed that mutant enzyme H373A significantly increased the yield of IMO4 to 62.32%. Saturation mutation of H373R and its neighboring site S374R also increased the yield of IMO4. Analysis of the enzymatic properties revealed that H373R had a lower optimum temperature and maintained high enzyme activity under alkaline conditions. The findings provide important theoretical basis for the future production of IMO4 and analysis of dextranase structure.

Summary: Non-digestible isomaltooligosaccharides (NDIMOS) are functional ingredients in food and beverages that provide health benefits through the metabolism of gastrointestinal microorganisms. This study synthesized NDIMOS using enzymes from Leuconostoc mesenteroides and confirmed their digestibility and ability to inhibit the formation of insoluble glucans. NDIMOS also promoted the growth of Bifidobacterium strains and inhibited the growth of Salmonella enterica serovar typhimurium, suggesting their potential use in food, beverages, and pharmaceuticals.

Digestibility of Bacillus firmus K-1 pretreated rice straw by different commercial cellulase cocktails

Summary: The study introduces an eco-friendly method of removing xylan from rice straw using xylanolytic Bacillus firmus K-1, showing that smaller particle sizes of rice straw give similar xylan removal efficiency. The pretreated rice straw exhibited higher digestibility and greater glucan conversion under enzymatic hydrolysis, with the extent of cellulose hydrolysis depending on the types and formulations of enzymes.

Preclinical Safety Assessment of Bacillus subtilis BS50 for Probiotic and Food Applications

Summary: In this study, the safety of Bacillus subtilis BS50 was assessed through preclinical analyses. The results showed that BS50 genome does not encode any known toxins, is susceptible to commonly prescribed antibiotics, and does not have any negative effects on the viability and permeability of human intestinal epithelial cells. Therefore, BS50 has a preclinical safety profile similar to commercial probiotic strains and poses little risk to human health.
