Eosinophils Correlate with Epithelial-Mesenchymal Transition in Chronic Rhinosinusitis with Nasal Polyps

Mingjie Wang et al.

Summary: The study revealed that in patients with eosinophilic chronic rhinosinusitis with nasal polyps, there is a downregulation of E-cadherin and an upregulation of Vimentin in epithelial tissue. The expression levels of Vimentin correlated with sinus CT imaging Lund-Mackay scores, numbers of infiltrating eosinophils in tissues, and peripheral blood eosinophil ratio. EMT occurred in CRSwNP patients, especially those with eosinophilic chronic rhinosinusitis with nasal polyps, suggesting a correlation between epithelial reprogramming, eosinophil infiltration, and disease severity. Eosinophils were found to contribute to impairment of epithelial function and promotion of EMT in CRSwNP.

Summary: This study investigated the role of TNF receptor-associated factor 6 (TRAF6) in the human neutrophil elastase (HNE)-induced autophagy and mucin MUC5AC over-expression in chronic rhinosinusitis with nasal polyps (CRSwNP). The results showed that HNE promotes autophagy through TRAF6, resulting in hyperexpression of MUC5AC in CRSwNP.

TIM-4 in macrophages contributes to nasal polyp formation through the TGF-β1-mediated epithelial to mesenchymal transition in nasal epithelial cells

Danxue Qin et al.

Summary: Chronic rhinosinusitis with nasal polyps (CRSwNP) is caused by prolonged inflammation of the paranasal sinus mucosa. In this study, TIM-4 was found to be increased in the sinonasal mucosa of CRSwNP patients, especially in macrophages. TIM-4 was positively correlated with clinical scores and inhibition of TIM-4 reduced polyp formation and EMT.

Mucin 5AC is significantly upregulated in exosomes from the nasal lavage fluid and may promote the expression of COX-2, VEGF and MMP-9: an implication in nasal polyp pathogenesis

Ling-Feng Wang et al.

Summary: Exosomes isolated from nasal lavage fluids of nasal polyps (NP) patients showed significantly upregulated mucin 5AC levels. These mucin 5AC-enriched exosomes were effectively taken up by fibroblasts derived from chronic rhinosinusitis without NP (CRSsNP) patients, leading to increased expression of COX-2, VEGF and MMP-9. Mucin 5AC carried by nasal exosomes may play a role in tissue remodeling and angiogenesis, making it a potential therapeutic target for NP.

Necroptosis Underlies Neutrophilic Inflammation Associated with the Chronic Rhinosinusitis with Nasal Polyps (CRSwNP)

Furthermore, DAMPs, inflammatory cytokines, and chemokines were increased in non-eosinophilic CRSwNP, and the extent of necroptosis was positively correlated with the abundance of these factors as well as neutrophil infiltration. Additionally, TNF-alpha and IFN-gamma induced necroptosis in THP-1 cells, suggesting a potential role in exacerbating inflammation in CRSwNP.
