The results showed that pretreatment with itaconate alleviated lung injury and inhibited LPS-induced lipid peroxidation and injury. Mechanistically, itaconate inhibited ferroptosis by regulating the accumulation and activation of Nrf2 and GPX4.

Necroptosis triggers spatially restricted neutrophil-mediated vascular damage during lung ischemia reperfusion injury

Summary: Ischemia reperfusion injury is a common pathological condition triggered by the release of endogenous ligands. In this study, the researchers discovered that necroptosis, a nonapoptotic form of cell death, triggers the recruitment of neutrophils in injured lungs. Initially, neutrophils traffic predominantly to subpleural vessels, guided by chemoattractants produced by nonclassical monocytes. The disruption of pulmonary capillaries by neutrophils leads to vascular leakage and impaired graft function. The study also found that TLR4 signaling and NADPH oxidase 4 expression mediate the arrest of neutrophils, preventing their extravasation. The formation of neutrophil extracellular traps in injured lungs can be disrupted with DNase to prevent vascular leakage and improve graft dysfunction. These findings could have important implications for the development of therapeutics to protect lungs from ischemia reperfusion injury.

Activation of ferritinophagy through a decrease in NRF2 and phosphorylation of ERK1/2 plays a key role in initiating ferroptosis in macrophages.

Circulating levels of DDIT4 and mTOR, and contributions of BMI, inflammation and insulin sensitivity in hyperlipidemia

It also identified decreased levels of DDIT4 and increased levels of mTOR in individuals with hyperlipidemia. Insulin resistance was positively correlated with blood pressure, BMI, lipid profiles, mTOR, and inflammatory markers, but negatively correlated with HDL-C and DDIT4. Lipid profiles were positively correlated with BMI, mTOR, and inflammatory markers, but negatively correlated with DDIT4. Factor analysis identified four domains related to hyperlipidemia.

Forsythoside A Mitigates Alzheimer's-like Pathology by Inhibiting Ferroptosis-mediated Neuroinflammation via Nrf2/GPX4 Axis Activation

Summary: Forsythoside A (FA), a main constituent of Forsythia suspensa, has been found to possess antioxidant, anti-inflammatory, and neuroprotective properties. This study investigated the potential role of FA in Alzheimer's disease (AD) and found that FA treatment improved mitochondrial function, inhibited lipid peroxidation, and reduced inflammation in AD-related cells and mice. Additionally, FA treatment attenuated memory and cognitive impairments, suppressed beta-amyloid deposition and tau hyperphosphorylation, and modulated dopamine signaling in the brain. These findings suggest that FA may have therapeutic potential as a drug component for AD treatment.

Quercetin alleviates acute kidney injury by inhibiting ferroptosis

Summary: The study demonstrated that quercetin (QCT), a natural flavonoid, inhibited ferroptosis in renal proximal tubular epithelial cells by reducing lipid ROS levels and increasing glutathione (GSH) levels. QCT also ameliorated acute kidney injury induced by ischemia-reperfusion (I/R) or folic acid (FA). Furthermore, RNA-sequence analysis revealed ATF3 as a key factor in QCT's inhibition of ferroptosis. Additionally, QCT inhibited the recruitment of pro-inflammatory macrophages in AKI.
