Assessment of Anti-inflammatory Activity of 3-Acetylmyricadiol in LPS-Stimulated Raw 264.7 Macrophages

Summary: This study isolated 3-Acetylmyricadiol, a pentacyclic triterpenoid, from the ethyl acetate bark extract of Myrica esculenta, and evaluated its anti-inflammatory potential. The results showed that 3-Acetylmyricadiol inhibited the production of nitric oxide, IL-6, and TNF-alpha in a dose-dependent manner, suggesting its potential as an effective anti-inflammatory molecule.

Modulation of proteasome activity by curcumin and didemethylcurcumin

Summary: Curcumin and its synthetic polyphenolic derivative, didemethylcurcumin, modulate proteasome activity in a biphasic manner. Curcumin increases proteasome activity at nanomolar concentrations but inhibits it at micromolar concentrations. Docking simulations show that curcumin has a stronger binding interaction with the proteasome compared to CUIII, suggesting their potential as therapeutic agents for neurodegeneration and cancer.

Extracted from marine sponges, over 5,000 marine natural compounds, including aaptamine, have been studied extensively and show promise for various biological purposes, such as cancer treatment and protease inhibition.

Summary: Curcumin is suggested to dysregulate the ubiquitin-proteasome system (UPS) and this study investigated the effects of curcumin supplementation on markers of UPS activity in response to muscle damage. The results showed that curcumin supplementation can regulate basal UPS activity during exercise-induced muscle damage.

Design, Synthesis, Molecular Modelling, and Biological Evaluation of Oleanolic Acid-Arylidene Derivatives as Potential Anti-Inflammatory Agents

Summary: In this study, a series of 15 new arylidene derivatives of oleanolic acid were synthesized by Claisen Schmidt condensation reaction. Among them, derivative 3L was identified as the most potent inhibitor of IL-6 and TNF-alpha, while compounds 3a, 3d, and 3e showed potent inhibition of NO. In addition, analogs 3e, 3L, and 3n exhibited greater affinity towards TNF-alpha and IL-1 beta than dexamethasone. These findings suggest that the synthesized compounds have promising anti-inflammatory potential.

Ubiquitin Proteasome Pathway Transcriptome in Epithelial Ovarian Cancer
