Lipogenesis inhibitors: therapeutic opportunities and challenges

Battsetseg Batchuluun et al.

Summary: Fatty acids play essential roles in cellular survival and function as bioenergetic substrates, structural components, and signaling molecules. Cells have evolved de novo lipogenesis (DNL) to generate fatty acids from alternative carbon sources. However, abnormal upregulation of DNL is associated with various pathologies. Inhibiting core enzymes of DNL represents a potential therapeutic strategy, and several synthetic DNL inhibitors are currently in development. Aberrant upregulation of DNL is implicated in conditions such as cardiovascular disease, metabolic disorders, and cancers.

Class I histone deacetylases (HDAC1-3) are histone lysine delactylases

Carlos Moreno-Yruela et al.

Summary: Lysine L-lactylation is a newly discovered histone modification that is stimulated under conditions of high glycolysis. Through systematic evaluation of histone deacetylases (HDACs), HDAC1-3 and SIRT1-3 were identified as delactylases for this modification. This study provides important insights into the regulatory mechanisms of histone lactylation.

Lactate promotes macrophage HMGB1 lactylation, acetylation, and exosomal release in polymicrobial sepsis

Kun Yang et al.

Summary: High levels of lactate and HMGB1 are associated with the severity and mortality of sepsis. Lactate can promote HMGB1 lactylation and acetylation in macrophages, leading to HMGB1 release and increased endothelium permeability. Targeting lactate-associated signaling pathways may offer a potential strategy to combat sepsis.

Positive feedback regulation of microglial glucose metabolism by histone H4 lysine 12 lactylation in Alzheimer's disease

Rui-Yuan Pan et al.

Summary: The dysregulation of microglial function in Alzheimer's disease is associated with the glycolysis/H4K12la/PKM2 positive feedback loop. Inhibiting this loop can ameliorate pathological changes and cognitive deficits in a mouse model. This suggests that disrupting the positive feedback loop may be a potential therapeutic approach for treating AD.

High Intensity Interval Training: A Potential Method for

Qian-Qi Liu et al.

Summary: Sarcopenia, an age-related disease, has severe impacts on the aging population. Traditional persistent aerobic exercise is being replaced by high intensity interval training (HIIT), which has shown similar or even better effects in improving muscle strength, physical performance, and muscle mass in older individuals. However, there is a lack of direct research on the effects of HIIT on sarcopenia patients.
