Terpenoid Transport in Plants: How Far from the Final Picture?

Olivia Costantina Demurtas et al.

Summary: Contrary to well-characterized biosynthetic pathways, the transport and secretion of terpenoids at different cellular and tissue levels are still poorly understood. This review aims to describe the current understanding of terpene transport and localization with a focus on physiologically and ecologically important terpenes. While only a small fraction of the thousands of identified terpenoids have been characterized for their transport, it is known that most are secreted in the apoplast or stored in vacuoles by ABC transporters. However, little is known about the movement of biosynthetic intermediates within the cell.

Metabolic Engineering of Crocin Biosynthesis in Nicotiana Species

Oussama Ahrazem et al.

Summary: In this study, the key enzyme CsCCD2L for producing high-value pigments called crocins was introduced into tobacco and wild tobacco plants. It was found that the transgenic wild tobacco plants accumulated higher levels of crocins compared to the transgenic tobacco plants. Furthermore, co-expression of CsCCD2L, BrCrtZ, and AtOrMut in tobacco plants resulted in a 3.5-fold increase in crocin production. These findings demonstrate the feasibility of producing saffron pigments in engineered Nicotiana species and provide a foundation for future strategies to commercialize these valuable pigments.

Summary: This study demonstrates the production of crocins in tobacco plants using BdCCD4.1 enzyme and highlights the higher accumulation of crocins in leaves compared to petals. The production of crocins is associated with decreased levels of beta-carotene and the stability of crocins decreases during long-term storage.

Summary: Molecular farming utilizes crop plants as biofactories to produce high value-added compounds, with Nicotiana species being one of the most suitable 'chassis' due to their nonfood status and high metabolic versatility. Using new plant breeding techniques, enriched Nicotiana species can be designed for molecular farming purposes.

Identification and characterization of apocarotenoid modifiers and carotenogenic enzymes for biosynthesis of crocins in Buddleja davidii flowers

Gianfranco Diretto et al.

Summary: The biosynthesis of crocetin in Buddleja davidii flowers involves a specific pathway catalyzed by carotenoid cleavage dioxygenases and subsequent reactions. Different crocins and carotenoid precursors accumulate in parallel with tissue development, with gene expression patterns mimicking crocin accumulation. Comparative analysis among different plant species highlights distinct differences in crocin accumulation and apocarotenoid synthesis.

Alberto Jose Lopez-jimenez et al.

Summary: Research has shown that crocetin is formed by carotenoid cleavage enzymes and specific CCD4 enzymes, accumulating in the stigma of saffron in the form of glucosides and crocins. UGT91P3 is believed to be the last missing gene in the biosynthesis of highly glucosylated crocins in saffron.
