Human Hematopoietic Stem Cell Engrafted IL-15 Transgenic NSG Mice Support Robust NK Cell Responses and Sustained HIV-1 Infection

Shawn A. Abeynaike et al.

Summary: Mice reconstituted with human immune systems are important for studying HIV-1. NK cells play a key role in innate anti-HIV responses, but current mouse models have limited NK cell development. Lack of human IL-15 signaling is a major obstacle. This study shows that NSG-Tg(IL-15) mice with human IL-15 expression support long-term engraftment of human NK cells.

Next-generation humanized NSG-SGM3 mice are highly susceptible to Staphylococcus aureus infection

Sophia Hung et al.

Summary: Humanized mice have shown increased vulnerability to Staphylococcus aureus infection compared to wild type mice, and this study found that next-generation humanized mice with improved myeloid reconstitution were even more susceptible. The severity of infection was correlated with the rate of humanization. These findings have implications for therapy approaches and analysis of virulence mechanisms.

Toward a humanized mouse model of Pneumocystis pneumonia

Guixiang Dai et al.

Summary: Recent genomic data suggest that divergence of major surface glycoproteins may play a role in speciation and host range selectivity of Pneumocystis fungus. Humanized mice provide information about human immune responses involved in controlling Pneumocystis infection. Experiments showed that CD34-engrafted huNOG-EXL mice were able to control fungal burdens more effectively and generate fungal-specific IgM.

High-throughput enrichment and isolation of megakaryocyte progenitor cells from the mouse bone marrow

Lucas M. Bush et al.

Summary: This study presents a high throughput strategy for identifying and isolating megakaryocytes and their progenitor cells, enabling rapid disease identification and diagnoses ahead of severe disease progression. By combining thrombopoietin induction, image flow cytometry, and principal component analysis, they have distinguished and enriched for megakaryocyte progenitor cells capable of self-renewal and directly differentiating into mature megakaryocytes, potentially revolutionizing disease research in this area.

IFN-γ-Stimulated Apoptotic Keratinocytes Promote Sclerodermatous Changes in Chronic Graft-Versus-Host Disease

Akimasa Saito et al.

Summary: In patients with GVHD, KCs in skin lesions of chronic GVHD show marked production of TGF beta 1, while IFN-gamma-deficient CD8 T-cell recipients have lower expression of TGF beta 1 in the epidermis. IFN-gamma promotes TGF beta 1 production by apoptotic KCs, mediating the development of widespread sclerodermatous changes in KC-targeting GVHD.

Teaching a new mouse old tricks: Humanized mice as an infection model for Variola virus

Christina L. Hutson et al.

Summary: Research has found that three humanized mice strains are highly susceptible to VARV infection, establishing the first small animal model. Following a VARV challenge mimicking human smallpox transmission, the virus spread systemically within the humanized mice similar to the course of human smallpox, facilitating testing of medical countermeasures.

Responses of Mast Cells to Pathogens: Beneficial and Detrimental Roles

Mariela Jimenez et al.

Summary: Mast cells are immune cells strategically located in tissues close to the external environment, playing a key role in the host defense against invading pathogens. They release biologically active mediators to induce inflammation, promote antimicrobial activities, and shape innate and adaptive immune responses.

Summary: Both graft-versus-host disease (GVHD) and inflammatory bowel disease (IBD) are immunologically mediated diseases of the intestine, sharing several key features in their onset and development including intestinal tissue damage and involvement of cytokines.

Innate Immune Reconstitution in Humanized Bone Marrow-Liver-Thymus (HuBLT) Mice Governs Adaptive Cellular Immune Function and Responses to HIV-1 Infection

Wilfredo F. Garcia-Beltran et al.

Summary: The study found that HuBLT mice have well-reconstituted T cells with intact diversity, but exhibit weak T-cell responses to HIV-1 challenge due to a deficiency in innate immune cells, particularly monocytes. Research suggests that efforts to improve the model for HIV-1 immune response and vaccine studies should focus on restoring innate immune reconstitution.
