Whole-genome sequencing data of Corynebacterium diphtheriae isolated from diphtheria outbreaks in Indonesia

Vivi Setiawaty et al.

Summary: Corynebacterium diphtheriae (C. diphtheriae) is the causative agent of diphtheria, and its main virulence factor is diphtheria toxin. In this study, whole-genome sequencing of C. diphtheriae isolates from diphtheria outbreaks in different regions in Indonesia was performed to identify single nucleotide polymorphisms (SNPs) associated with the tox and dtxR genes. Molecular typing using multilocus sequence typing (MLST) was also conducted. The data obtained can be used for further analysis of antimicrobial resistance and bacterial virulence factors.

Fatal Respiratory Diphtheria Caused by β-Lactam-Resistant Corynebacterium diphtheriae

Brian M. Forde et al.

Summary: In this study, a fatal case of respiratory diphtheria caused by toxigenic C. diphtheriae resistant to penicillin and all other beta-lactam antibiotics was reported. A novel mechanism of inducible carbapenem resistance associated with the acquisition of a mobile resistance element was also described.

Assessing the Genetic Diversity of Austrian Corynebacterium diphtheriae Clinical Isolates, 2011 to 2019

Justine Schaeffer et al.

Summary: Diphtheria is a vaccine-preventable disease with high potential for reemergence. A study in Austria found high genetic diversity among isolated C. diphtheriae strains, with some carrying antimicrobial resistance genes. Whole-genome sequencing provided valuable information for identifying clusters, evaluating antimicrobial resistance, and supporting toxigenicity results obtained by PCR. Routine implementation of whole-genome sequencing could greatly benefit C. diphtheriae surveillance.

Molecular and Epidemiological Characterization of Toxigenic and Nontoxigenic Corynebacterium diphtheriae, Corynebacterium belfantii, Corynebacterium rouxii, and Corynebacterium ulcerans Isolates Identified in Spain from 2014 to 2019

Andreas Hoefer et al.

Summary: This study analyzed toxigenic and nontoxigenic Corynebacterium isolates submitted to the national reference laboratory in Spain between 2014 and 2019. The results showed a predominance of Corynebacterium diphtheriae and Corynebacterium belfantii, with 7 new sequence types described for the first time in Spain. Some isolates carried antibiotic resistance genes, and patients with toxigenic isolates had adverse outcomes.

Summary: This study identified key predictors of mortality in pediatric patients with clinical diphtheria, including immunization status, clinical features, and laboratory profiles. The presence of these features upon hospital admission must be taken into account, as they can lead to fatal outcomes.

Epidemiological, clinical, and public health response characteristics of a large outbreak of diphtheria among the Rohingya population in Cox's Bazar, Bangladesh, 2017 to 2019: A retrospective study

Jonathan A. Polonsky et al.

Summary: The background unrest in Myanmar in August 2017 led to the displacement of over 700,000 Rohingya refugees to crowded camps in Cox's Bazar, Bangladesh, resulting in a large outbreak of diphtheria. Data from mass vaccination campaigns, contact tracing, and treatment centers were used to describe the epidemiological and clinical features of the outbreak. With 7,064 reported cases between November 2017 and November 2019, the outbreak mainly affected children with a low case fatality ratio. High population density, poor living conditions, and fast growth rates contributed to the rapid spread of the outbreak, with mass vaccination campaigns showing modest reductions in transmission.

Genomic Epidemiology and Strain Taxonomy of Corynebacterium diphtheriae

Julien Guglielmini et al.

Summary: The study aims to understand the phylogeography and short timescale evolution of Corynebacterium diphtheriae using a core genome multilocus sequence typing (cgMLST) scheme, identifying sublineages and strain tracking in outbreaks. The high allele call rate and genomic differences among strains demonstrate the effectiveness of cgMLST for epidemiological surveillance and public health management of diphtheria. The unified genomic taxonomy provided by the study offers a common language for studies of ecology, evolution, and virulence heterogeneity among C. diphtheriae sublineages, enhancing our understanding of the pathogen's transmission dynamics and geographical spread.
