Current considerations for fetal alcohol spectrum disorders: identification to intervention

Leila Glass et al.

Summary: This review summarizes recent findings on the prevalence, public health impact, clinical presentation, intervention access, and conceptualization of fetal alcohol spectrum disorders (FASDs). Despite ongoing efforts in prevention and identification, the rates of drinking during pregnancy have increased, and there are significant gaps in diagnosis and intervention.

Fetal alcohol spectrum disorders

Svetlana Popova et al.

Summary: Fetal alcohol spectrum disorders (FASD) are a group of conditions characterized by neurodevelopmental impairment with or without facial dysmorphology, congenital anomalies and poor growth. This Primer provides an overview of FASD, including epidemiology, pathophysiology, diagnosis, and treatment. Prenatal alcohol exposure (PAE) can have harmful effects on fetal development, and the risk is influenced by various factors such as dose, pattern, timing, and duration of exposure, as well as genetic and environmental factors. FASD is a significant public health issue with profound social and economic consequences, but it often goes undiagnosed or receives inadequate attention.

Prenatal exposure to alcohol impairs responses of cerebral arterioles to activation of potassium channels: Role of oxidative stress

Partha S. Saha et al.

Summary: Prenatal alcohol exposure can lead to dysfunction in the ability of specific potassium channels to dilate cerebral arterioles, potentially due to increased oxidative stress. These alterations may contribute to the pathogenesis of cerebral vascular abnormalities and behavioral/cognitive deficits observed in fetal alcohol spectrum disorders.

Reducing Prenatal Alcohol Exposure and the Incidence of FASD: Is the Past Prologue?

Grace Chang

Summary: This narrative review summarizes and synthesizes the clinical trials and randomized clinical trials that evaluated selected and targeted approaches to reducing preconception and prenatal alcohol exposure (PAE) and alcohol-exposed pregnancy (AEP) since 2011. The search results identified 21 studies that focused on case management, preconception efforts, motivational interviewing and screening, brief intervention, and referral to treatment, as well as the use of technology to deliver the intervention. The findings suggest that case management and home visits may not be effective in reducing alcohol exposure, while preconception efforts and technological approaches show promising potential.

Blood brain barrier as an interface for alcohol induced neurotoxicity during development

Michele Siqueira et al.

Summary: Ethanol consumption during pregnancy or lactation permanently impairs the development of the central nervous system, resulting in fetal alcohol spectrum disorders (FASD). FASD encompasses a range of deficits in the embryo caused by alcohol exposure during gestation, with fetal alcohol syndrome (FAS) being the most severe. FAS is characterized by facial abnormalities, stunted growth, low body weight, and structural and/or functional damage to the central nervous system. Ethanol has deleterious effects on different brain regions, leading to microcephaly, abnormal cortical development, corpus callosum agenesis or hypoplasia, and cognitive and behavioral deficits. Teratogenic ethanol particularly affects the CNS blood vessels, which constitute the neurovascular unit of the blood-brain barrier (BBB). BBB disruption can worsen neurodegenerative and neurological diseases.

Development and Validation of a GC-EI-MS/MS Method for Ethyl Glucuronide Quantification in Human Hair

Alessandro Mattia et al.

Summary: Ethyl glucuronide (EtG) is a minor ethanol metabolite that can be detected in various matrices. This study developed and validated a new method for analyzing EtG in hair samples using gas chromatography and tandem mass spectrometry. The method was successfully applied to real hair samples and used to assess excessive alcohol consumption and verify the psycho-physical requirements of individuals.

The Status of Oxidative Stress in Patients with Alcohol Dependence: A Meta-Analysis

Mi Yang et al.

Summary: Alcohol dependence leads to oxidative stress in the body, as evident from the changes observed in the levels of oxidative stress biomarkers. This meta-analysis highlights the decreased activities of superoxide dismutase (SOD) and glutathione peroxidase (GPx) enzymes and lower levels of albumin, along with increased levels of malondialdehyde (MDA), vitamin B12, homocysteine, and bilirubin in patients with alcohol dependence.

Ginevra Micangeli et al.

Summary: Oxidative stress is a condition characterized by an imbalance between antioxidant and oxidative factors. It has serious consequences on the body and is related to various pediatric syndromes, either as a main etiological factor or directly associated with genetic alterations. Further research is needed to better understand the relationship between oxidative stress and pediatric diseases.

Summary: Research has shown that neonatal alcohol exposure may lead to learning and memory deficits as well as depressive behavior in adult rats, associated with increased oxidative stress parameters in the hippocampus and prefrontal cortex. The complete reversal of these effects by Rapamycin suggests its effectiveness in preventive therapy for disorders related to prenatal alcohol exposure.
