Characterization of Vemurafenib-Resistant Melanoma Cell Lines Reveals Novel Hallmarks of Targeted Therapy Resistance

Martina Radic et al.

Summary: Despite the advancements in melanoma treatment, many patients still develop resistance to drugs, such as the RAF inhibitor vemurafenib. In this study, researchers generated and examined two melanoma-derived cell lines with acquired resistance to vemurafenib. They found that the resistant cells displayed features of epithelial-mesenchymal transition (EMT) and exhibited a hybrid phenotype. Surprisingly, the resistant cells showed decreased migration ability but had a tendency for collective migration. Signaling pathway analysis revealed reactivation of MAPK and activation of the PI3K/AKT pathway in the vemurafenib-resistant cells. The acquired resistance also led to resistance to chemotherapy in the primary melanoma cells. Further investigation is needed to understand the negative regulators of NME proteins in vemurafenib resistance and their impact on PI3K/AKT signaling and melanoma cell characteristics.

Phytochemical profile and biological activities from different parts of Vaccinium vitis-idaea

Di Zhang et al.

Summary: This study evaluated the active ingredients, antioxidant, and enzyme-inhibitory activity of different parts of wild lingonberry. Phenolic compounds were found to be significantly higher in the leaf and stem. Fruit exhibited the highest inhibitory activity on acetylcholinesterase and butyrylcholinesterase, while the leaf showed the most potent activity on alpha-amylase, alpha-glucosidase, and tyrosinase. Tyrosinase was strongly correlated with phenolic acids and flavonoids. A total of 29 active ingredients, including phenolic acids, flavonoids, anthocyanins, and triterpenes, were detected. The results suggest that lingonberry has potential as a natural resource for functional food and medicinal development.

Composition, sun protective and antimicrobial activity of lipophilic bilberry (Vaccinium myrtillus L.) and lingonberry (Vaccinium vitis-idaea L.) extract fractions

Linards Klavins et al.

Summary: Berries of the genus Vaccinium found in bogs and forests of Northern Europe are a valuable source of biologically active substances containing sterols, unsaturated fatty acids, flavonoids, and anthocyanins, which provide various health benefits such as treating urinary tract inflammation, controlling blood sugar levels, improving cardiovascular health, and reducing oxidative stress caused by free radicals. Studies have shown that lipid fractions of bilberry and lingonberry contain fatty acids, sterols, isoprenoids, and long-chain aliphatic compounds, and that fractions with high concentrations of phenolic acids have potential UV-B blocking activity, while those with high sterol content effectively inhibit bacterial growth. These findings are important for the development of natural functional ingredients for nutraceuticals and cosmeceuticals.
