Methylation of melatonin receptors in patients with unipolar and bipolar depression

Summary: Disturbances in melatonin secretion and its receptors MT1 and MT2 are observed in depressed patients, affecting their circadian rhythm and sleep-wake cycle. This study aimed to evaluate the methylation profile of melatonin receptor genes MTNR1A and MTNR1B in peripheral blood mononuclear cells (PBMC) of depressed patients. The methylation pattern differed in depressed patients, with lower methylation levels of MTNR1A compared to healthy controls. Moreover, melatonin concentration was negatively correlated with MTNR1B methylation level in the depressed patients. These findings suggest that the methylation profile of melatonin receptors in PBMC may serve as a complementary molecular marker in depression diagnosis.

Summary: Breast cancer, a common cause of cancer-related death in women, requires further studies and revolutionary treatment approaches. Epigenetic alterations in normal cells are strongly associated with the development of breast cancer, and targeting these alterations through epigenetic-based therapy shows promise. Specific enzymes, such as DNA methyltransferases and histone deacetylases, play crucial roles in the formation and maintenance of epigenetic changes and are potential targets for treatment. Epidrugs target different epigenetic alterations and can restore normal cellular memory in cancerous diseases. This review focuses on the important role of epigenetic regulation and the clinical implications of epidrugs in breast cancer.
