Synthesis of New Shogaol Analogues as NRF2 Activators and Evaluation of Their Anti-Inflammatory Activity, Modes of Action and Metabolic Stability

6-Shogaol inhibits the proliferation, apoptosis, and migration of rheumatoid arthritis fibroblast-like synoviocytes via the PI3K/AKT/NF-κB pathway

Summary: This study found that 6-shogaol inhibits proliferation, migration, and invasion, and induces apoptosis in rheumatoid arthritis fibroblast-like synoviocytes (FLSs). It also reduces the production of inflammatory cytokines and matrix metalloproteinases (MMPs). Molecular mechanism analysis revealed that 6-shogaol inhibits the PI3K/AKT/NF-kappa B pathway by activating PPAR-gamma. Treatment with 6-shogaol ameliorated joint destruction in a mouse model of collagen-induced arthritis (CIA). Therefore, 6-shogaol may be a potential natural drug for future rheumatoid arthritis treatments.

6-Shogaol Exhibits Anti-viral and Anti-inflammatory Activity in COVID-19-Associated Inflammation by Regulating NLRP3 Inflammasomes

Summary: Recent global health concern has led to the investigation of natural medicinal plant resources as alternative treatments for COVID-19 infection and inflammation. In this study, a phytochemical called 6-shogaol was explored as a potential anti-inflammatory and anti-COVID-19 agent. 6-shogaol was found to efficiently inhibit SARS-CoV2 replication and regulate the expression of inflammasome pathway molecules. Compared to existing drugs, 6-shogaol demonstrated similar efficacy and better safety in terms of physicochemical properties and clinical toxicity. This study provides important insights for the further development of 6-shogaol as a potential therapeutic agent against inflammation and viral infections.

Metabonomics and the gut microbiome analysis of the effect of 6-shogaol on improving obesity

Summary: This study investigated the effects of 6-shogaol on reducing obesity in C57BL/6J mice fed with a high-fat diet. After 12 weeks of intervention, it was found that 6-shogaol reduced obesity levels and blood lipid levels in the mice, and improved glucose homeostasis, especially at low doses. The intervention also influenced the gut microbiota and the metabolic profile of the mice. The findings suggest that 6-shogaol may have potential in improving obesity and provide insight into its molecular mechanisms.
