Hepcidin and Iron in Health and Disease

Elizabeta Nemeth et al.

Summary: Hepcidin, secreted by hepatocytes, controls the activity of ferroportin to regulate plasma iron concentrations and total body iron content. Iron loading and inflammation increase hepcidin levels, while erythropoietic stimulation and pregnancy suppress its levels. Hepcidin deficiency leads to iron overload, while hepcidin excess causes iron-restrictive anemias. Developing hepcidin diagnostics and therapeutics will improve the treatment of iron disorders.

Modulation of the HIF2α-NCOA4 axis in enterocytes attenuates iron loading in a mouse model of hemochromatosis

Nupur K. Das et al.

Summary: This study found that NCOA4-mediated intestinal ferritinophagy is integrated into systemic iron demand via HIF2 alpha. Disruption of intestinal NCOA4 did not affect the intestinal and systemic response to iron deficiency, but provided protection against iron overload in a mouse model.

Iron-induced NCOA4 condensation regulates ferritin fate and iron homeostasis

Sota Kuno et al.

Summary: NCOA4 acts as a crucial autophagy adaptor protein that regulates ferritin fate under both iron repletion and depletion conditions, maintaining cellular iron homeostasis. By forming insoluble condensates, NCOA4 prevents excessive iron storage in cells and facilitates ferritin delivery to lysosomes, effectively balancing iron levels in the cell.

Protective Effects of Hif2 Inhibitor PT-2385 on a Neurological Disorder Induced by Deficiency of Irp2

Jiaqi Shen et al.

Summary: The upregulation of Hif2 alpha in tissues of Irp2(-/-) mice, particularly in the central nervous system, was confirmed in this study. Inhibition of Hif2 alpha prevented neurodegenerative symptoms, suggesting its potential as a clinical target in the treatment of IRP2 deficiency-induced neurodegenerative syndrome.

CD63 is regulated by iron via the IRE-IRP system and is important for ferritin secretion by extracellular vesicles

Izumi Yanatori et al.

Summary: Extracellular vesicles (EVs) transfer functional molecules between cells, with CD63 expression regulated by the IRE-IRP system in response to iron levels. Under iron loading, cellular ferritin is transferred via NCOA4 to CD63(+) EVs for secretion, providing insight into the local exchange of ferritin and iron between cells.
