Near-physiological in vitro assembly of 50S ribosomes involves parallel pathways

Summary: We investigated the assembly principles of 50S ribosomal subunits using cryo-EM and found that they assemble based on cooperative assembly blocks and defined dependencies. We resolved 13 intermediate structures that accumulate during synthesis, revealing the smallest known assembly core. Our findings shed light on the parallel pathways involved in the early and late assembly stages of the 50S subunit.

MINFLUX dissects the unimpeded walking of kinesin-1

Summary: We introduce an interferometric MINFLUX microscope that can record protein movements with up to 1.7 nanometer per millisecond spatiotemporal precision. It only requires the detection of about 20 photons from a 1-nanometer-sized fluorophore, eliminating the need for attaching large beads to the protein. With this microscope, we were able to study the stepping of kinesin-1 on microtubules at physiological ATP concentrations, revealing rotations of the stalk and heads of the protein during stepping.

Direct observation of motor protein stepping in living cells using MINFLUX

Summary: We developed a live-cell tracking method with nanometer spatial and millisecond temporal resolution using MINFLUX super-resolution technique. This method allowed us to resolve the precise stepping motion of motor protein kinesin-1 on microtubules in living cells. Tracking of motors on fixed cell microtubules also revealed the architecture of the microtubule cytoskeleton with protofilament resolution.

Cryo-EM captures early ribosome assembly in action

Summary: Ribosome biogenesis is a multi-step cellular process that involves the production, processing, folding, and modification of ribosomal RNAs (rRNAs) and ribosomal proteins. By using a minimal in vitro reconstitution system, researchers studied the early assembly phase of the bacterial 50S subunit and found that it occurs in a domain-wise fashion, with late assembly proceeding incrementally. The study also showed that ribosomal proteins and folded rRNA helices induce or stabilize rRNA folds within adjacent regions, creating cooperativity.
