Summary: This study investigated the role of Lgr4/5 in nonalcoholic fatty liver disease (NAFLD) and found that the loss of Lgr4/5 in hepatic epithelial cells led to hepatic steatosis and fibrosis in mice fed with a high-fat diet. The study also demonstrated that the loss of Lgr4/5 resulted in impaired bile acid secretion, altered lipid homeostasis, and deregulation of lipoprotein pathways, all of which contributed to the onset of NAFLD.

Accelerating Ozonolysis Reactions Using Supplemental RF-Activation of Ions in a Linear Ion Trap Mass Spectrometer

Summary: Gas-phase ion-molecule reactions provide valuable structural insights in analytical applications. However, the slow reaction rates limit their widespread use. In this study, a supplementary RF-activation method is used to accelerate the rate of ion-molecule reactions involving ozone. The accelerated reaction rates increase sample throughput and enable identification of low abundance lipid isomers, expanding the range of lipid species that can be analyzed.

Branched and linear fatty acid esters of hydroxy fatty acids (FAHFA) relevant to human health

Summary: Fatty acid esters of hydroxy fatty acids (FAHFAs) are a complex lipid class that play important roles as signaling mediators and structural components. Two structurally and functionally distinct FAHFA superfamilies are recognized based on the position of the estolide bond. The combination of different hydroxy fatty acids and fatty acids, along with the position of the estolide bond, generates a wide variety of unique FAHFA structures. This review highlights the anti-diabetic and anti-inflammatory effects of branched FAHFAs, the role of omega-FAHFA-derived lipids in tear film lipid layer and dry eye disease, and provides insights into the biosynthesis, degradation pathways, and distribution of FAHFAs in adipose tissue.

ATGL is a biosynthetic enzyme for fatty acid esters of hydroxy fatty acids

Summary: In this study, the researchers identified adipose triglyceride lipase (ATGL) as the first biosynthetic enzyme for the formation of the FAHFA ester bond in mammals. They found that ATGL uses transacylase activity to esterify hydroxy fatty acids (HFAs) with fatty acids (FAs) from triglycerides (TGs) or diglycerides (DGs) to produce FAHFAs. The expression of ATGL and the activity of DGAT were found to be closely associated with FAHFA biosynthesis.

ATGL liberates FAHFAs preferentially from more compact substrates, while HSL acts as a potent estolide bond hydrolase for both TAG estolides and free FAHFAs.
