Hydrogen sulfide and epigenetics: Novel insights into the cardiovascular effects of this gasotransmitter

Summary: Epigenetics studies heritable modifications of genome expression, which do not affect the DNA sequence. Alterations in epigenetic mechanisms can impact phenotype and contribute to disease. Recent evidence suggests that hydrogen sulfide (H2S) plays a role in epigenetic regulation, from DNA methylation to histone modifications and non-coding RNA regulation. This review explores the potential for developing H2S-releasing "epidrugs" for the prevention and treatment of cardiovascular and non-cardiovascular disorders based on its regulatory role in epigenetics.

The Impact of Drugs on Hydrogen Sulfide Homeostasis in Mammals

Summary: Mammalian cells and tissues have the ability to produce hydrogen sulfide gas (H2S) through cysteine metabolism. H2S plays roles in cell signaling cascades essential in many biochemical and physiological processes in various organs and systems of mammals. Reduced levels of H2S are observed in several pathophysiological conditions, and it has been discovered that certain commonly prescribed drugs can affect the enzymes responsible for H2S production. Therefore, this review summarizes the studies on key drugs and their impact on H2S production in mammals.

CBS and its product hydrogen sulfide (H2S) play various roles in tumor growth, cell protection, and angiogenesis in high-CBS expressing tumor cells. This article reviews the potential of CBS silencing and pharmacological CBS inhibition as potential cancer therapies, and discusses the identification of biomarkers for future targeted treatments.

Their beneficial cardiovascular effects are attributed to the release of hydrogen sulfide (H2S) in biological tissue, which induces vasorelaxation. H2S acts on various molecular targets to promote vasodilation, including opening of potassium channels, inhibition of 5-phosphodiesterase, and activation of VEGFR-2. These compounds have novel mechanisms of action and potential as antihypertensive agents.
