Shotgun knockdown of RNA by CRISPR-Cas13d in fission yeast

Summary: The CRISPR-Cas13d system is a promising tool for RNA manipulation, as it targets RNA with a small effector protein and does not require specific sites in the target transcript. In this study, the CRISPR-Cas13d system was successfully implemented in fission yeast for reversible RNA knockdown. The system's effectiveness was ensured by using an array of CRISPR RNAs (crRNAs) expressed from an RNA polymerase II promoter. This new toolkit is expected to have diverse applications in biology, including gene function analysis and RNA visualization.

The collateral activity of RfxCas13d can induce lethality in a RfxCas13d knock-in mouse model

Summary: The knockdown of gene expression using RfxCas13d in adult brain neurons results in the death of mice, which is likely due to the collateral activity of RfxCas13d rather than the loss of target gene function or off-target effects. Mechanistically, RfxCas13d exhibits collateral activity in mammalian cells, which is positively correlated with the abundance of target RNA. The collateral activity of RfxCas13d cleaves 28s rRNA into two fragments, leading to translation attenuation and activation of the ZAK alpha-JNK/p38-immediate early gene pathway.

An improved auxin-inducible degron system for fission yeast

Summary: Conditional degron technologies, such as the auxin-inducible degron (AID) system, are important tools for biological research. In this study, we improved the AID system for the fission yeast Schizosaccharomyces pombe by optimizing all three components: the AID degron, the small-molecule inducer, and the inducer-responsive F-box protein. Our improved AID system showed inducible lethality for 10 essential genes, including ones that could not be effectively inactivated using a previously published AID system.
