Selectively Targeting Breast Cancer Stem Cells by 8-Quinolinol and Niclosamide

Summary: This study aimed to find potential drugs targeting cancer stem cells (CSCs) in triple negative breast cancer (TNBC). The results showed that 8-quinolinol (8Q) and niclosamide (NCS) exhibited significant anti-CSC activity in vitro and in vivo, and had a synergistic effect with PTX in inhibiting the proliferation of cancer cells and viability of CSCs. Additionally, the combination of NCS and PTX reduced tumor growth and metastasis. These findings provide new insights for the development of more effective treatments for TNBC.

Cancer Stem Cells: Biology and Therapeutic Implications

Morphological features of single cells enable accurate automated classification of cancer from non-cancer cell lines

Summary: This research successfully introduced an algorithm that could distinguish cancer from non-cancer cells in high-resolution single cell images by exploiting structural differences in the cytoskeleton. Overcoming the barrier of insufficient data in machine learning methodologies, the study identified the most discriminatory features to predict cell type accurately with as few as 100 cells per line. Furthermore, normalizing cell shape improved discrimination of cell lines with difficult-to-distinguish phenotypes, demonstrating the generalizability of the algorithm across various tissue origins.

Identification of MFGE8 and KLK5/7 as mediators of breast tumorigenesis and resistance to COX-2 inhibition

Summary: Our study identified multiple COX-2 associated genes that play important roles in tumor lung colonization in triple negative breast cancer, with silencing of MFGE8 and KLK5/7 genes significantly restoring sensitivity to COX-2 selective inhibitor in TNBC cells both in vitro and in vivo. This suggests that novel COX-2 inhibitor-based combination therapies could be effective strategies for future TNBC treatment.
