Mitochondrial DNA variation across 56,434 individuals in gnomAD

Kristen M. Laricchia et al.

Summary: This study presents a pipeline to call mtDNA variants and applies it to the gnomAD v3.1 database, reporting population frequencies for a large number of mtDNA variants, which will be helpful for clinical interpretation and research studies.

Mito-SiPE is a sequence-independent and PCR-free mtDNA enrichment method for accurate ultra-deep mitochondrial sequencing

Darren J. Walsh et al.

Summary: The analysis of somatic variation in the mitochondrial genome requires deep sequencing of mitochondrial DNA. This study presents a PCR-free method for ultra-deep sequencing coverage of the mitochondrial genome, using isolated intact mitochondria and a sequence-independent approach. The method avoids false-heteroplasmy calls caused by long-range PCR amplification and enables researchers to identify low frequency heteroplasmy without introducing PCR biases or NUMT contamination.

NUMTs Can Imitate Biparental Transmission of mtDNA-A Case in Drosophila melanogaster

Maria-Eleni Parakatselaki et al.

Summary: This article investigates the inheritance of mtDNA in fruit flies and reveals mtDNA heteroplasmy caused by NUMT interference. The study highlights the need to exclude the possibility of NUMT interference in mtDNA inheritance studies.

Nuclear-embedded mitochondrial DNA sequences in 66,083 human genomes

Wei Wei et al.

Summary: The transfer of mitochondrial DNA into the nucleus contributes to a complex landscape of nuclear-mitochondrial segments (NUMTs). Almost all individuals have multiple NUMTs, including rare ones that are present in less than 0.1% of the population. Most NUMTs inserted into the nuclear genome after humans diverged from apes. Once inserted, the sequences are no longer under mitochondrial evolutionary constraints and have different mutational signatures compared to mitochondrial DNA.

A bioinformatics pipeline for estimating mitochondrial DNA copy number and heteroplasmy levels from whole genome sequencing data

Stephanie L. Battle et al.

Summary: This article presents a bioinformatics pipeline for identifying mitochondrial DNA variations and quantifying mitochondrial copy number, which more accurately captures the genetic features of mitochondria. The pipeline improves accuracy through methods such as remapping data, recovering reads, and using consensus chrM sequences.

Inhibition of mitochondrial and cytosolic calpain attenuates atrophy in myotubes co-cultured with colon carcinoma cells

XianLiang Zeng et al.

Summary: Cancer cachexia is characterized by muscle atrophy and mitochondrial injury, which can be induced by calpain activation. In this study, calpain inhibitors were used to prevent muscle atrophy by inhibiting calpain activity in myotubes during co-culture with carcinoma cells. Additionally, these inhibitors improved mitochondrial function in myotubes and prevented calpain-induced muscle wasting during co-culture.

Mitochondrial Transfer Improves Cardiomyocyte Bioenergetics and Viability in Male Rats Exposed to Pregestational Diabetes

Eli J. Louwagie et al.

Summary: Offspring born to diabetic or obese mothers have a higher lifetime risk of heart disease. Research has shown that exposure to late-gestational diabetes mellitus (LGDM) or maternal high-fat (HF) diet can lead to mitochondrial dysfunction, impaired cardiomyocyte bioenergetics, and cardiac dysfunction in rat offspring. Findings suggest that mitochondrial dysfunction may play a central role in neonatal cardiomyopathy, with sex-specific differences observed in mitochondria-mediated mechanisms of heart disease development. Therapeutic mitochondrial transfer may have potential limitations, such as inconsistent effects in male and female offspring.
